| There are a large number of dead animals and poultry in China every year,and the meat of dead animals and poultry contains rich organic resources.The pollution-free treatment of dead livestock and poultry is the key problem in the treatment of dead livestock and poultry.The utilization of biodegradable livestock and poultry meat can effectively improve the utilization rate of resources,reduce environmental pollution and waste of organic resources.High temperature liquid hydrolysis of dead livestock and poultry by high temperature bacteria functional strains can control the temperature artificially,which can not only effectively kill pathogenic microorganisms,but also obtain nutrient rich liquid organic fertilizer.In this experiment,the microorganisms in the soil of high temperature environment were enriched and cultured at 70 ℃ and the high temperature protease and lipase resistant functional strains were screened.Six strains were obtained by plate screening,shaking bottle screening,enzyme activity determination and single strain hydrolysis of meat.Among them,the protease activity of the crude enzyme solution of strain H1 with the best hydrolysis effect was 23.33 U/m L and lipase activity was 11.99 U/m L.Through morphological observation,physiological and biochemical experiments and molecular biological identification,strains H1,H12 and H13 were identified as Geobacillus stearothermophilus,The strain Db1 is Geobacillus subterraneus,Db48 is Geobacillus thermophiliovans,and H16 is Geobacillus kaustophilus.Different strains were used to hydrolyze pork,duck and chicken with 5% addition at 70 ℃ respectively.After 5 days of hydrolysis,strain Db1 had the highest degree of hydrolysis of pork,which was 85.82%.The content of water-soluble protein,small peptide,amino acid and total free amino acid in pork hydrolysate inoculated with strain H1 were10.563 mg/m L,9.362 mg/m L and 1.927 mg/m L,respectively.Strain H1 had the highest degree of hydrolysis of duck meat,which was 89.15%.The content of soluble protein,small peptide,amino acid and total free amino acid in duck hydrolysate inoculated with strain H1 were 9.125mg/m L,8.990 mg/m L and 1.640 mg/m L,respectively.Strain Db48 had the highest degree of hydrolysis,which was 88.52%.The content of water-soluble protein,small peptide and amino acid in chicken hydrolysate inoculated with strain H1 were 8.596 mg/m L and 8.501mg/m L,respectively.The total amount of free amino acids in chicken hydrolysate inoculated with strain Db35 was the highest,which was1.867 mg/m L.The mixed broth containing strains Db1,Db48,H1,H12,H13 and H16 was hydrolyzed at high temperature for different meats with 5%,10%,15%,20% and 25%,and the optimal addition amount and hydrolysis time of meat were explored.Considering the degree of hydrolysis,the content and yield of water-soluble protein,small peptide,amino acid and free amino acid,the optimal adding amount of pork is10% and the optimal hydrolysis time is 84 h,under these conditions,the degree of hydrolysis of pork is 85.53%,and the content of water-soluble protein is 24.217 mg/m L,the yield of water-soluble protein was 92.57%,the content of small peptide and amino acid was 22.765 mg/m L,the yield was 86.49%,the content of free amino acid was 2.825 mg/m L,the yield was 10.73%.Under these conditions,the degree of hydrolysis was85.26%,the content of water-soluble protein was 38.856 mg/m L,and the yield of water-soluble protein was 80.36%;the content of small peptide and amino acid was 36.756 mg/m L,the yield of small peptide and amino acid was 76.01%;the content of free amino acid was 6.199 mg/m L,the yield was 12.82%.Under these conditions,the hydrolysis degree of chicken was 91.69%,the content of water-soluble protein was 61.869mg/m L,the yield of water-soluble protein was 86.45%;the content of small peptide and amino acid was 54.869 mg/m L,the yield of small peptide and amino acid was 77.17%;the content of free amino acid was8.494 mg/m L,the yield of water-soluble protein was 11.95%.. |
