| Seed is one of the most basic means of production in agricultural production,and its quality directly affects the yield and quality of crops.Seed priming technology is proposed based on the biological mechanism of seed germination,which aims to promote seed germination,increase the stable rate of germination time and germination regularity,and improve seedling resistance and nutritional status.This experiment uses stale Chinese cabbage seeds stored at room temperature for 5 years as materials,and uses reagents such as γ-poly-glutamic Acid(γ-PGA),5-aminolevulinic acid(5-ALA),γ-aminobutyric acid(GABA)and compound sodium nitrophenolate for single factor treatment and orthogonal combination treatment.The seed germination,seedling quality and antioxidant enzyme activities of Brassica campestris L.under different ratio treatments and different simulated stress conditions were survey and evaluate,the suitable ratio of initiator was selected and used in germination test of old seeds of watermelon,onion and tomato.Verify its applicability to different seeds and provide a strong theoretical basis for the selection of seed initiators.The main findings are as follows:(1)Through a single factor test,it was found that 100 mg/L~500 mg/L γ-PGA treatment,10 mg/L~30 mg/L 5-ALA treatment,5 mmol/L~20 mmol/L GABA treatment and 1 mg/L~10 mg/L compound sodium nitrophenolate,the treatments all promoted the germination of seeds,and the germination index,growth index and antioxidant enzyme activity of seeds were significantly better than the control treatment.(2)The experiment found that T9 treatment with different combination ratios: 500 mg/L γ-PGA,10mg/L 5-ALA,10 mmol/L GABA,10 mg/L compound sodium nitrophenolate reagent combination triggering effect was significantly better than Other combinations.At this time,the germination index and root length of Chinese cabbage seeds were increased by 46.40%,24.62%,31.51%,45.68% and 10.77% respectively compared with T0 treatment;the soluble protein content and the activities of POD,SOD and CAT were also significantly increased.In addition,the T9 treatment combination had a certain promoting effect on the germination and seedling growth of old seeds of watermelon,onion and tomato.(3)Under drought and salt stress treatments,although the germination index of seeds treated with T9 is still lower than the germination index after initiation treatment under no stress treatment,the germination potential,germination rate,germination index and germination index of seeds are improved to a certain extent.Vitality index,its various germination indexes are higher than untreated germination indexes under no stress conditions;the soluble protein,MDA content and antioxidant enzyme activity in seedlings are increased compared with the untreated level.To sum up,the T9 treatment screened in this study through the screening of a single initiator and the combination of different reagents can promote the germination of old Chinese cabbage seeds;increase the soluble protein content and antioxidant enzyme activity in the seedlings.Thereby alleviating the damage of adversity stress to seed germination and improving the resistance of seedlings. |
