| Tobacco(Nicotiana tabacum L.)seed is an irreplaceable means of production in tobacco production.Hunan Province is one of the main tobacco producing areas in China,but at present,Yongzhou City in Hunan Province is facing the following major problems:(1)the germination rate and vigor of tobacco seeds are relatively low,and the uniformity of seedling emergence is relatively poor;(2)the technology of seed coating pelletization is relatively backward,which makes the heavy metal content in coated seeds,leading to relatively serious seed borne diseases;(3)the technology of seed quality detection is not advanced。Therefore,on the basis of the existing tobacco production technology,taking Xiangyan 5 and GX-8 as the experimental materials and Yunyan 87 as the control,this study carried out the determination of the seed vigor of naked and coated seeds,the improvement of the scheme of soaking and germination of naked seeds,and the improvement of coating technology.In order to provide help for tobacco seed coating technology in Yongzhou City,Hunan Province.The main results are as follows:1.Detection of tobacco naked seed vigorArtificial accelerated aging for 2 days can distinguish the first,second,GX-8 hybrid and third,fourth and fifth grades of Xiangyan 5 by vigor index,which can make the difference between low vigor seeds and high vigor seeds reach a significant level.In the experiment of low temperature naked seed germination,only the varieties or grades with different vigor can be distinguished,but it is more difficult for the more refined vigor grades.2.Detection of seed vigor of tobacco coated seedsAfter 2 days of artificial accelerated aging,the germination index and average germination days of seeds were analyzed,and the three varieties were distinguished.The activity of antioxidant enzymes and the accumulation of MDA in seedlings were closely related to the level of seed vigor.The level of seed vigor could be preliminarily determined by detecting them.At the same time,it was found that the germination rate,average germination days,germination index and dry weight of the three varieties could reach the significant difference level when they were differentiated in low temperature.3.Tobacco seed germination scheme(1)The germinating time of Xiangyan No.5 was 24 hours and 24 hours respectively when it was germinating with GA3 100mg/L,and 48 hours and 24 hours respectively when it was germinating with negative ion Powder 100g/L;(3)the germinating time of GX-8 was 24 hours and 24 hours respectively when it was germinating with GA3 100mg/L.When the negative ion powder was 100g/L,the two times of germination were 48h and 12h,respectively.4.Tobacco coating scheme and supporting technology Xiangyan No.5 and GX-8 were pelletized with the improved germinating scheme and the new and old coating materials,and compared with the coating seeds of the old materials and the non improved germinating scheme.After that,the standard germination experiment,artificial accelerated aging experiment and low temperature germination experiment were carried out to test the germination index,vigor index and physiological index.The final pelletization scheme was determined as follows;(1)Xiangyan 5 was treated with GA3100mg/L as germination promoter,the two times of germination were 24 hours,and the coating material was treated with new material(2)GX-8 was treated with negative ion powder 100 g/L was used as a germinating agent for 24 hours,and the coating material was a new material. |
