| Scutellaria baicalensis Georgi is the herb perennials which belongs to Scutellaria under Lamiaceae,Laliatae.It has the functions such as clearing heat,drying dampness,antipyretic,detoxification,haemostasis,protecting the fetus and so on.It’s widely used in clinical in China.And Chinese doctor used it as a common medicinal materials for a long time.The seeds of it are small and mainly direct sow.So it is difficult to germinate.If the soil is dry,the topsoil is uneven or the soil particles are large when sowing,it will be difficult to emergence and then lead to the phenomenon of lack of seedling.At present,there is no national standard for Scutellaria baicalensis Georgi seeds.This study explored the seed quality testing methods of Scutellaria baicalensis Georgi and established the quality classification criteria of it.Meanwhile,the Hydropriming and Hormonal priming technologies were applied to the process of seed germination under salt stress.The main results were as follows:1.The quality of Scutellaria baicalensis Georgi seeds from five Northern areas in China(Hebei,Shanxi,Gansu,Shandong and Shaanxi)were measured based on the International seed testing regulations and the crop-seed testing regulations.The quality inspection methods were established as follows.Using the microscope observation method to test the appearance and authenticity of Scutellaria baicalensis Georgi seeds.The samples were at least 50 g when measure purity and at least 5 g for one test.The thousand kernel weight was measured by 500-seed method.The water content was tested by low temperature drying method after grinding and sieving: 75℃±2℃ for 8 hours.Seed viability was tested by TTC methods: after soaking in distilled water for 12 hours,peel off the seed coat and use 0.5%TTC dyeing at 25℃ for 12 h.The germination rate was determined by paper germination experiment: 25 ℃,the incubator whose relative humidity is 60%,and the counting time was 2-8 days.2.In this part,we selected Eighteen batches of Scutellaria baicalensis Georgi from different producing areas in total.And the seed purity,thousand kernel weight,water content,seed viability and seed germination rate were measured and analyzed through SPSS20.0 software by K cluster analysis.Considering the actual production and operability,We used the seed germination rate and water content as the main index for classification.While the seed viability,thousand kernel weight and purity were used for important references.The quality classification criteria was divided into three grades as following: for the first grade,germination rate≥70%,seed viability≥70%,moisture content≤6%,seed purity≥85%,thousand kernel weight≥1.9 g;for the second grade,germination rate≥50%,seed viability≥50%,moisture content≤7%,seed purity≥80%,thousand kernel weight≥1.8 g;for the third grade,germination rate≥30%,seed viability≥30%,moisture content≤8%,seed purity≥75%,thousand kernel weight≥1.7 g.3.The effect of different priming time,different concentrations of GAs priming and different concentrations of 6KT priming were studied on the germination rate,seedling biomass,root length and seedling height of Scutellaria baicalensis Georgi.And the non-priming seeds was used as a control.The results showed that the best priming time is24 hours,and the optimal condition of GAs is 300 mg/L for 24 hours,the optimal condition of 6KT is 5 mg/L for 24 hours.The suitable conditions of priming which advanced the initial germination time point by shortening the mean germination time can improve the germination performance of Scutellaria baicalensis Georgi.Meanwhile,the seedlings can germinate fast and concentrated.In addition,GAs and 6KT can also promote the elongation of roots and seedlings.4.The effect of the best priming time,the optimal condition of GAs and 6KT under salt stress were studied on germination and the activities of peroxidase,superoxide dismutase and catalase.Also using the non-priming seeds as a control.The results showed that the germination of Scutellaria baicalensis Georgi was significantly inhibited under Na Cl stress.Seed germination rate was significantly higher than the control group after hydropriming and Hormonal priming.As Na Cl concentration rising,the activities of peroxidase,superoxide dismutase and catalase trended downward.But after priming in suitable conditions,the activities of those three enzymatic of seedlings of Scutellaria baicalensis Georgi enhanced significantly compared to the control group. |
