The purpose of this experiment was to use Congo red medium and carboxymethyl cellulose sodium cellulose(CMC-Na),microcrystalline cellulose,and D-salicylic medium as the sole carbon source,respectively,on the cellulose-degrading bacteria in the intestinal tract of bamboo rat.Carry out primary screening,study the enzyme-producing activity of the strains obtained from the primary screening under different culture conditions,and select a cellulose-degrading bacteria with higher enzyme activity,aspergillus niger and Enterococcus faecalis as single or mixed bacteria Form solid-state fermentation of Moringa oleifera stalks,select the best fermentative strain combination based on the content of crude protein,crude fiber,neutral detergent fiber,acid detergent fiber,and crude fat in the fermented Moringa oleifera stalks,and use the response surface methodology.The fermentation conditions were optimized to improve the nutritional value of Moringa stalks.The results show: The purpose of this experiment was to use Congo red medium and carboxymethyl cellulose sodium cellulose(CMC-Na),microcrystalline cellulose,and Dsalicylic medium as the sole carbon source,respectively,on the cellulosedegrading bacteria in the intestinal tract of bamboo rat.Carry out primary screening,study the enzyme-producing activity of the strains obtained from the primary screening under different culture conditions,and select a cellulosedegrading bacteria with higher enzyme activity,aspergillus niger and Enterococcus faecalis as single or mixed bacteria Form solid-state fermentation of Moringa oleifera stalks,select the best fermentative strain combination based on the content of crude protein,crude fiber,neutral detergent fiber,acid detergent fiber,and crude fat in the fermented Moringa oleifera stalks,and use the response surface methodology.The fermentation conditions were optimized to improve the nutritional value of Moringa stalks.The results show:(1)A total of 11 strains of bacteria were isolated from the intestinal tract of bamboo rat.After the primary screening of the degrading bacteria in Congo red medium,five strains of cellulose-degrading bacteria,N1,N2,N3,N4,and N5,were obtained;the enzyme activities were determined,and N3 The activities of endoglucanase(Cen),exoglucanase(Cex),β-glucosidase(BG)and filter paper enzyme(FPA)were significantly higher than those of N1,N2,N4 and N5 strains(P <0.05),the filter paper enzyme(FPA)activity of five strains was determined under different culture conditions,and N3 was also better than N1,N2,N4,and N5 strains;after identification,N3 strain was Bacillus subtilis.(2)After fermentation,the content of crude protein and crude fat in the three bacteria combination T7 group formed by Bacillus subtilis + Aspergillus niger + Enterococcus faecalis was significantly higher than that in the control group CK and single bacteria or double bacteria group T1,T2,T3,T5,T6groups(P﹤0.05),there was no significant difference compared with T4 group(P﹥0.05);the crude fiber content in T7 group was significantly lower than that in CK,T1,T2,T3,T5,T6 groups(P﹤0.05)Compared with the T4 group,there was no significant difference(P﹥0.05);the content of acid detergent fiber in the T7 group was significantly lower than that in the CK,T1,T2,T3,T4,T5,T6groups(P﹤0.05),and the neutral detergent fiber content was significantly lower It was lower than that of CK,T1,T2,and T3 groups(P﹤0.05),and compared with T4,T5,and T6 groups,the difference was not significant(P﹥0.05).(3)The response surface methodology was used to optimize the solid-state fermentation conditions of Moringa oleifera stems.The model predicted that the optimal fermentation conditions were water content of 70%,fermentation temperature of 37.7degrees Celsius,inoculum size of 14%,and fermentation time of 11.4 days.After fermentation,the crude protein content can be increased to 16.58%,and the crude fiber content can be reduced to 35.28%.After the verification test,the model is more reliable.To sum up,the best strain of cellulose-degrading bacteria isolated from the intestinal tract of bamboo rat is N3,and after identification,N3 is Bacillus subtilis;Bacillus + Aspergillus niger + Enterococcus faecalis;the optimal fermentation conditions were optimized by response surface methodology: water content of 70%,fermentation temperature of 37.8degrees Celsius,inoculum amount of 14%,and fermentation time of 11.4 days.
