| Pregnancy toxemia in goat is a serious nutritional metabolic disease that occurs frequently in elderly and prolific ewes.The disease is prone to the death of animals.and its pathogenesis is still unclear.In this study,we detected the biochemical changes of the blood in goats,and analyzed the changes of serum metabolome in goats with pregnancy toxemia.To provide a research basis for further analysis the pathogenesis of goat pregnancy toxemia.Methods: Nine goats with suspected pregnancy toxemia(PT group)and nine healthy goats during the same period of pregnancy(NC group)were selected for this study.the serum was collected when the disease happens,and liquid chromatography-tandem mass spectrometry(LC-MS/MS)technology was used to compare the serum metabolite changes in PT and NC groups.The main results are as follows:1.Serum biochemical indexes showed that aspartate aminotransferase(AST)and β-hydroxybutyrate(B-HB)were significantly increased,albumin(ALB)was significantly decreased,glucose and calcium ion were extremely significantly decreased in PT group compared with NC group.There are no significant difference on other biochemical indexes between these two group.2.Serum metabolomics identified a total of 5236 metabolites in positive ion mode,of which 1588 were differential metabolites,724 were up-regulated and 864 were downregulated.Significant differential metabolites such as palmitoylcarnitine,L-tryptophan,Citric acid,α-ketoglutaric acid,chenodeoxycholic acid;1782 metabolites were identified in negative ion mode,of which 560 were differential metabolites,260 were up-regulated,and 300 were down-regulated.Significant differential metabolites such as L-tyrosine Acid,cholic acid,glycocholic acid,L-proline.3.Pathway enrichment analysis showed that 35 differential metabolite pathways were enriched in positive ion mode,and 7 differential metabolic pathways were enriched in negative ion mode.The significantly different metabolites are mainly enriched in cholesterol metabolism pathway,bile secretion pathway,primary bile synthesis pathway,phenylalanine,tyrosine and tryptophan biosynthesis pathway,amino acid biosynthesis pathway.Conclusion: The serum metabolism of ewes changes significantly during the onset of goat pregnancy toxemia.Differential analysis of new metabolites helps to unravel goat pregnancy toxemia pathogenesis. |
