Paeonia suffruticosa is native to China and has been cultivated there for more than a thousand years.Peony is a woody plant.It takes 3~5 years from sowing to flowering,and longer time to apply for new variety right after the characters are stable.There are a certain proportion of self-bred progenies in the hybrid progenies,so it is very important to identify the authenticity of the seedling of the hybrid progenies in the early stage,which is helpful to improve the breeding efficiency and avoid unnecessary waste of resources in the later stage.In this study,we used ’Bao Qing Hong’ and ’Feng Dan’ as parents to hybridize with Japanese tree peony cultivars with high double-petaled.We identified and analyzed 35 hybrids from 6 cross combinations,speeding up the breeding process of excellent varieties of tree peony.The main findings are as follows:(1)’Bao Qing Hong’ and ’Feng Dan’ were used as parents in the cross breeding with Japanese peony varieties.It was found that they had high cross bearing ability.The hybrid combination of ’Bao Qing Hong’ × ’Ba Qian Dai Chun’,’Bao Qing Hong’×’Hua Jing’,’Dao Da Chen’ × ’Bao Qing Hong’,’Dao Jin’ × ’Bao Qing Hong’,’Feng Dan’ × ’Dao Jin’,’Hua Jing’ × ’Feng Dan’ has high seedling emergence rate and strong seed growth vigor.(2)By observing and comparing 24 morphological indexes of leaf length,leaf color,lateral leaflet,flower type,flower color,flower disc and carpel between parents and offspring,it was preliminarily determined that there were 35 hybrids,27 of which were true hybrids,and 9 of them were partial paternal inheritance.It was found that some of the hybrids were abortive,and the hybrids with new phenotypes need to be further analyzed.(3)The 7 pairs of SSR primers with good polymorphism and clear amplified bands were screened from 125 pairs of SSR primers,and M1-2,M1-5,M2-3,M2-5,M2-7,M2-9,M4-1,M4-3,M5-1,M5-2 10 true hybrid progenies were identified with complementary bands specific to their parents,it is a new germplasm resource of Paeonia suffruticosa.(4)The Hybrid M5-4 had parental morphological characteristics and was judged to be a true hybrid,but there were no parental characteristics in SSR molecular marker analysis,so it should be a false hybrid.The results showed that morphological identification was subjective and needed to be combined with other identification methods to make more accurate identification.In this study,morphological identification and SSR molecular markers were used to identify the hybrids of Tree Peony.The 7 pairs of primers can be used to identify the progenies of cross between Paeonia Suffruticosa and Paeonia Suffruticosa,and provide a theoretical reference for molecular breeding in later stage.
