| China is the largest potato producer in the world,but the per unit yield is lower than the world average.The main reason is the species degradation caused by the accumulation of virus generations when potato is propagated asexually by tuber.The breeding of virus-free seed potato is an effective measure to solve the degradation of seed potato.The modern seed potato production system is based on test-tube plantlets or microtuber to produce minituber,and then produce commercial seed potatoes at all levels.The purpose of this study is to optimize the production technology of microtuber production,to increase the weight of microtuber,to directly replace minituber,so as to reduce the production process.The experimental results showed that the application of blue light treatment could effectively promote the induction of potato in vitro.The main results are as follows:1.Compared with traditional white light LED culture conditions,blue light LED culture conditions were used to induce microtuber.The results showed that blue light treatment had obvious promoting effect on tuber formation.2.On this basis,the effect of blue light intensity on the formation of microtuber was systematically studied.40μmol·m-2·s-1,80μmol·m-2·s-1 and 120μmol·m-2·s-1 were set up respectively,and three potato varieties Zhongshu No.5,Huashu No.1 and Huashu No.9 were set up.The purpose of this study was to explore the most suitable conditions for blue light-induced tuber formation in each variety.The results showed that Zhongshu No.5 and Huashu No.1 had the best induction effect under 120μmol·m-2·s-1 blue light intensity,while Huashu No.9 had the same induction effect under three blue light intensity.Overall,after blue light treatment,the plant height of potato test-tube plantlets decreased,the stem diameter increased,the leaf area increased,and the stomatal development was promoted;the potato weight of microtuber increased,the proportion of effective potato increased,and the proportion of large potato increased;jasmonic acid synthesis and metabolic pathway were activated,and StSWEET11 gene expression in source-sink pathway was up-regulated.3.Zhongshu No.5 was used as the experimental material,and the blue light intensity of 120μmol·m-2·s-1 was set,which was combined with carbon dioxide,long sunshine and temperature difference respectively,in order to expect the effect of superposition on the induction of microtuber.The results showed that CO2 treatment alone could promote the induction effect of microtuber,and the promoting effect could be superimposed when treated with blue light at the same time.Long sunshine treatment alone could also promote the induction effect of microtuber,but when treated with blue light at the same time,the promoting effect could not be superimposed.The induction of microtuber treated with temperature difference alone was not as good as that of blue light alone,but when treated with blue light alone,the effect of tuber could be improved to be consistent with that of blue light alone. |
