There is bleeding phenomenon in many plants,which has an important impact on plant growth and development.And the bleeding phenomenon of Actinidia arguta particularly serious.A.arguta,also known as soft jujube,is a large deciduous vine belonging to A.arguta in kiwifruit family.It is rich as wild resources in Northeast China,but there are relatively few studies on its bleeding phenomenon.Therefore,this experiment was used A.arguta as materials to collect the bleeding sap of annual branches,biennial branches and bare branches.The bleeding law and main components were analyzed.The impact of bleeding on leaves,stems and fruits,in order to lay a theoretical basis for mastering the bleeding law of A.arguta and the effective application of bleeding sap.These main results are as followed:1.The results showed that the total amount of bleeding sap of biennial branches was higher than that of annual branches and bare branches.There were significant differences in DPPH·,O2·-,NO2-clearance,soluble sugar and soluble protein content of annual branches in different branches.The gibberellin content from high to low was:bare branches,annual branches and biennial branches.2.GC-MS was used to analyze the metabolomics of annual branch bleeding sap.Different metabolites were screened out.These metabolites included to amino acids(17%),organic acids(3%),sugars(16%),amines(6.5%),Polyols(6.5%),lipids(13%)and others(38%).In addition,the main metabolic pathways involved in glycolysis pathway,tricarboxylic acid cycle pathway,urea cycle,amino acid metabolism,lipid metabolism and so on.3.The anatomical structures of the main veins and stems of the leaves were observed by paraffin section technique.These results showed that the parenchyma cells and collenchyma of the main veins of the leaves were unevenly arranged and the structure was damaged by bleeding.After bleeding,the area of phloem,phloem fiber and xylem decreased,and the arrangement and distribution were irregular.4.Through the detection and analysis of physiological indexes of plants after occuring bleeding,the contents of malondialdehyde,soluble sugar,soluble protein and proline in leaves and stems increased from the initial flowering stage to the fruit setting stage.The contents of malondialdehyde,soluble sugar,soluble protein and proline decreased from fruit ripening stage to defoliation stage.ROS accumulated significantly in leaves and stems at full flowering stage and fruit ripening stage after bleeding,indicating that the occurrence of bleeding damaged the plant during growth and development.5.Through the morphological observation of plants in different growth stages,compared with the control group,the leaf area decreased after bleeding,and the number,length and new leaves of the stem decreased.In severe cases,parts of the plant died.In addition,the fruit appeared late ripening and stunting.Compared with the control group,the contents of vitamin C,soluble sugar and soluble solid in treatment group decreased after bleeding.It shows that bleeding had a great influence on fruit quality.
