Skin wound is one of the common problems in veterinary clinic. In order to accelerate skin wound healing, wound dressing,as a temporary skin substitute, creates an effective barrier to protect the wound from external environmental infection. However, the current research on wound dressings is no longer satisfied with the isolation of the external environment,so there is an increasing demand for efficient new dressings. The application of hydrogel dressings in wound repair has attracted extensive attention.Hydrogel dressings can provide a moist environment for the wound, and can efficiently pack and release the drug slowly, which is conducive to wound healing. In addition, chitosan has good biocompatibility, blood compatibility, mechanical compatibility and other characteristics, can promote the regeneration of wound tissue,as a biomedical material for skin wound repair has been widely concerned. Therefore,in this study,C57BL/6J mice were used as research objects to repair mouse skin wounds with chitosan hydrogel dressings of different concentrations, and to explore the efficiency of chitosan hydrogel in wound closure. The results showed that chitosan hydrogel dressing could fully adhere, fix and heal the wound, significantly shorten the wound healing time and accelerate the wound healing rate, and there was a certain difference in the wound healing speed between male and female mice.In addition, the expression of tissue repair related factors near the wound was detected by immunofluorescence and the wound skin pathological section was observed. The results showed that Chitosan hydrogel dressing can not only enhance the expressions of vascular endothelial growth factor(VEGF),epidermal growth factor(EGF),matrix metalloproteinase-9(MMP-9)and estrogen receptor-α(ER-α)around the wound, but also promote the expressions of apoptosis factors caspase-3,Bcl-2 and P53 around the wound. These results suggest that chitosan may affect the wound healing process in mice by affecting inflammation, granulation tissue proliferation, collagen fiber production and epidermal regeneration. In conclusion, chitosan hydrogel dressings can accelerate wound healing in mice, and provide a reference for the development of veterinary clinical drug dressings that can promote wound healing.
