Effect Of Compound Traditional Chinese Medicine Preparations On Wound Repair Of Bacterial Infection In Rabbits

Abstract/Summary:
In the veterinary clinic,skin wounds caused by factors such as fights and car accidents are more common.Trauma repair is constrained by various factors such as infection,licking,and tearing,which can cause delayed repair of trauma,making it difficult for veterinary clinical treatment.Modern medical treatment of trauma is effective,but there are problems such as high cost and safety.Traditional medicine has a long history and experience in treating trauma,and has a high level of safety,which makes up for the problems of modern medicine in treating trauma.In this experiment,we investigated the role of topical compound Chinese medicine preparations in promoting the repair of bacterial infected wounds in rabbits and its mechanism to provide a theoretical basis for veterinary clinical use of Chinese medicine preparations in the treatment of surgical wounds.The experiment combined the traditional Chinese veterinary theory,with the principle of using heat-clearing and detoxifying,antibacterial and anti-inflammatory drugs as the main drugs and blood-stasis activating,decaying and muscle-generating drugs as the auxiliary drugs in the formula.New Zealand white rabbits were used to establish a defective wound on the back using surgical instruments,and then a bacterial infection model was constructed by adding Staphylococcus aureus bacterial solution drop by drop to infect the wound.After the infection model was established,the infected wounds were grouped according to the herbal treatment group,Chuangfu Ling group,saline group and blank group and treated with drugs respectively,with Chuangfu Ling as the positive drug group.The effect of the infected wounds and the calculation of wound healing rate,inhibition test,peripheral blood platelet and leukocyte count,protein content in serum and trabecular granulation tissue,hydroxyproline content in serum and trabecular granulation tissue,serum IL-6 and the pathological histology of trabecular tissues after drug administration were observed to further explore the mechanism of action of the compound Chinese medicinal preparation to promote trabecular repair.The results showed that:(1)Compounded herbal preparations were applied to traumatic surfaces:after the drug was applied to the traumatic surface,the granulation tissue grew vigorously,the trauma edge was flatter,and the scar tissue was light after repair.The wound healing rates of the herbal treatment group and the Chuangfu Ling group differed significantly from those of the saline group on the 7th and 14th days after medication(P<0.05).(2)The compound Chinese herbal preparation could reduce bleeding,improve the phagocytosis of leukocytes,and enhance the local immune function of the wound:on the3th and 14th days after drug administration,the peripheral blood platelet counts of the Chinese herbal treatment group and the Chuangfu Ling group differed significantly from those of the saline group(P<0.05).On the 7th day after medication,the number of peripheral blood leukocytes in the herbal treatment group and the Chuangfu Ling group differed significantly from that in the saline group(P<0.05).(3)The compound Chinese herbal preparation could promote granulation tissue proliferation and accelerate the synthesis of fibroblasts and collagen:on the 7th day after medication,the levels of serum and trabecular granulation tissue protein and hydroxyproline in the Chinese herbal treatment group differed significantly from those in the saline group(P<0.05).(4)HE staining showed that the degree of inflammatory cell infiltration in the herbal treatment group and the Chuangfu Ling group decreased with the time of medication compared with the saline group,and the epithelial growth was intact and the fibroblasts and collagen fibers were densely arranged after wound repair.(5)The compound Chinese medicine preparation could reduce the serum IL-6 level and avoid excessive inflammatory reaction:on the 7th day after medication,the serum IL-6level in the Chinese medicine treatment group differed significantly from that in the saline group(P<0.05).The compound Chinese herbal preparation could accelerate the repair of bacterial infected wounds by increasing the protein and hydroxyproline content in rabbit serum and trabecular granulation tissue and decreasing the serum IL-6 content,etc.Compared with the Chuangfu Ling group,the Chinese herbal treatment group could promote the repair of wounds and significantly reduce the scar growth after the repair of wounds.
