Zhou 8425 B is the seed of Guangmai 74(Triticale)/Lianfeng 1//Piedmont Wheat hybridized by Zhoukou Academy of Agricultural Sciences with 60Co-γ gamma radiation,and then crossed with Annong 7959 to obtain a new translocation line of 1BL/1RS.The germplasm has the outstanding advantages of dwarf lodging resistance,large spike and large grain,disease resistance and resistance.Zhou8425 B provided new backbone germplasm and gene resources for wheat genetic breeding.1.In this study,the 1BL/1RS translocation fragment of Zhou 8425 B was analyzed,and the second generation sequence was used to sequence Zhou 8425 B,Yamanabe,Dwarf Meng and Fengdecunmai 5.Specific molecular markers of 8425B1BL/1RS translocation lines were developed based on the sequencing results and detected in 222 derivatives of 8425 B.From 1BL/1RS translocation fragments of Zhou 8425 B,476 In Del sequences with length of more than 10 bp were screened,of which 45 were specific In Del markers.Among them,3 pairs(ZH84-1,ZH84-42 and ZH84-74)could identify Zhou 8425 B from wheat varieties such as Lankao 908,Lankao 906,Lankao Paizao 8,Lovlin 10,Avril and Caucasus wheat.PCR amplification was carried out on 222derivatives(lines)of 8425 B using these three specific gene markers,and the positive rate was 34%.2.the resistance of 222 Zhou 8425 B derivatives to stripe rust was identified by single strain 34(CYR34)of virulence physiology of stripe rust.The resistance of hybrid strain CYR34 was used to identify the resistance of the derivatives to stripe rust.The results showed that Zhou 8425 B had high resistance to stripe rust to virulent dominant strain CYR34 at seedling stage and mature stage,and Zhou 8425 B carried resistance genes of Yr ZH84(all stage resistance),Yr ZH84.2(all stage resistance),Yr ZH22(adult plant resistance)and Yr30(adult plant resistance),which ensured that the resistance remained stable since 1988.Among the 222 varieties of 8425 B,there were 14 varieties with stable growth period and resistance,accounting for 6.3%;52 varieties with stable growth period and resistance,accounting for 23.4%.Zhou8425 B many derivatives mainly through Zhou Mai 11,Zhou Mai 12,Zhou Mai 13,Zhou Mai 15,Zhou Mai 16 and Zhou Mai 17 6 sub-generation derived to the second generation.The excellent stripe rust resistance gene of Zhou 8425 B is continuously isolated and polymerized during the process of genetic breeding.The frequencies of Yr ZH84,Yr ZH84.2,Yr ZH22,and Yr30 in the derived offspring are 34.7%,14.9%,66.7%,and42.3%,respectively.Among the derivative varieties carrying only one disease resistance gene,the average severity of carrying Yr ZH84 was the lowest(23.5%),and the average severity of carrying Yr30 was the highest(69.6%).The average severity of carrying Yr ZH22 and Yr ZH84.2 was 34.4%and 68.4%,respectively;Among the derivatives that aggregate two disease resistance genes,the average severity of carrying Yr ZH84+Yr ZH22 was the lowest(13.3%),while the average severity of carrying Yr ZH22+Yr30 was the highest(53.8%);Among the derived varieties that aggregate three disease resistance genes,the average severity of carrying Yr ZH84+Yr ZH22+Yr30 was the lowest(17.2%),and the average severity of carrying Yr ZH84.2+Yr ZH22+Yr30 was the highest(42.9%).The research results provide genetic information for the continuous improvement and utilization of wheat core germplasm Zhou 8425 B in China,and identify a new derived germplasm that exhibits resistance and high yield to the highly virulent physiological race CYR34,providing a reference for the genetic breeding of wheat resistance to stripe rust in China.
