Study On The Mechanism Of Influence Of Egg Quality Difference On Hatchability Of Huainan Partridge Chickens

Abstract/Summary:
The hatching effect was affected by egg quality.In order to explore the effect of egg quality on hatching rate and its regulation mechanism,800 40-week-old Huainan partridge chickens were selected in this study.The hatching rate of individual eggs was recorded and divided into high and low hatching rate groups(HH and LH groups).50 eggs of high and low hatching rate groups were collected for egg quality analysis.The magnum and uterus related to egg quality differences were collected for quantitative analysis of gene expression.Some uterus and magnum were sliced for structural analysis.Part of the uterus were taken for transcriptome sequencing analysis,and the expression of key proteins in the uterus was detected by Western Blot.The hatching rate of HH group was 100%,which was significantly higher than that of LH group(81.52%).The eggshell thickness and eggshell strength of HH group were significantly lower than those of LH group(P<0.05),while the egg weight,protein height and Haugh unit were significantly higher than those of LH group(P<0.05).There was no significant difference in dry matter,crude protein and fat content in egg white and yolk between the two groups(P>0.05).The weight of ovary and magnum,and the index of the magnum in HH group were significantly higher than those in LH group(P<0.05).The villus gap in HH group were significantly lower than those in LH group,but the epithelial cell height of uterus were significantly higher than those in LH group,(P<0.05).The expression of GALNT9 in HH group was significantly higher than that in LH group.Compared with LH group,a total of 1077 differentially expressed genes were detected in HH group,of which595 were up-regulated and 482 were down-regulated.GO enrichment analysis showed that DEGs were mainly enriched in transmembrane transport(GO:0055085)and ion transport(GO:0006811).KEGG enrichment analysis showed that DEGs were mainly enriched in Regulation of actin cytoskeleton,ECM-receptor interaction and Calcium signaling pathway.The candidate genes related to eggshell synthesis were CALB1,KCNJ16,SLC26A9 and SLC4A4.The expression of CALB1 protein in HH group was significantly higher than that in LH group,while the expression of SLC26A9 protein was significantly lower than that in LH group(P<0.05).In summary,the up-regulation of GALNT9 in the swelling part of HH group promoted the formation of glycoprotein in egg white,and the protein height and Haugh unit increased significantly.The candidate genes related to eggshell synthesis were CALB1,KCNJ16,SLC26A9 and SLC4A4.In the process of eggshell synthesis,the ion precursor HCO3-played an important role,and the expression of SLC26A9 protein in LH group was high,the eggshell thickness and eggshell strength increased significantly,and the hatching rate decreased.
