Acer truncatum,a deciduous tree species of the acer family,is china’s only economic tree species.Flavonoids,tannins,phenolic acids,lignans,and other active components are abundant in Acer truncatum leaves tablets.They have anticancer,anti-inflammatory,lipid-lowering,and antibacterial properties,and have a wide range of applications.At the moment,the active components of Acer truncatum leaves can be extracted quickly and efficiently using organic solvents,although there has been little research on the extraction method of flavonoids.Flavonoids are unstable due to reactions and oxidative degradation during usage and storage.As a result,this study extracts total flavonoids from Acer truncatum leaves using ultrasound assisted ethanol extraction and finds the best extraction method using single factor and response surface optimization experiments.The wall material was maltose dextrin(MD)andβ-Cyclodextrin(β-CD),and spray drying was used to prepare the microencapsulation of total flavonoids of Acer truncatum leaves,in order to improve the stability of total flavonoids of Acer truncatum leaves and provide technical reference for the industrial development of Acer truncatum leaves.The following are the key research findings:(1)The best extraction process settings for total flavonoids from Acer truncatum leaves are 1:30 g/m L material to liquid ratio,10 minutes ultrasonic time,60%ethanol concentration,60°C ultrasound temperature,and 300 W ultrasound power.The yield of total flavonoids is 7.05%under these conditions,while the purity of flavonoids in the extract is 51.37%.The total flavonoid extract of Acer truncatum leaves includes670 components,including flavonoids,tannins,phenolic acids,lignans,and coumarins,according to liquid chromatography-mass spectrometry.(2)The best process conditions for spray drying microcapsules are MD:β-CD=1:1(w/w),solid concentration 20%(w/w),core to wall ratio 1:4(w/w),inlet temperature160°C;under these conditions,the embedding rate of total flavonoids is 70.58%,and the powder extraction rate is 56.7%.The microcapsule is a delicately textured light yellow powder.(3)Acer truncatum leaves total flavonoids physicochemical properties the retention rate of total flavonoids and DPPH free radical scavenging capability were used as markers to study the storage stability of microcapsules.Microcapsules have a moisture content of 2.72%,a solubility of 48.59%,a stacking density of 0.41 g·cm-3,a compaction density of 0.58 g·cm-3,and a wettability of 14.43 minutes.During the 28-day storage period,the retention rate of total flavonoids and DPPH radical scavenging ability of microcapsules was about 10%higher than that of the extract under dark,low temperature(20°C),and vacuum conditions;under three storage conditions of light,high temperature(60°C),and air exposure,the total flavonoid retention rate and DPPH radical scavenging capacity retention rate of microcapsules were still significantly higher than those of extract.Microencapsulation can reduce total flavonoid loss,protect total flavonoid storage stability,and greatly improve total flavonoid storage stability.The ultrasonic aided extraction technique and microcapsule fabrication process of total flavonoids from Acer truncatum leaves were optimized in this study,and the composition of the total flavonoids extract from Acer truncatum leaves was identified and studied.Microcapsules can efficiently preserve and stabilize total flavonoids.The findings of the study give theoretical and technological references for the extensive processing and high-value exploitation of Acer truncatum leaves.
