Auricularia heimuer belongs to the Fungi,Basidiomycota,Auriculariales,Auriculariaceae,Auricularia heimuer,is a kind of wood-rotting fungi growing in medium-temperature environment.Since the 1980 s,the polyethylene bag used to cultivate Auricularia heimuer successfully,it has been widely used in production in main producing in world’s leading producer.Production volume perennial ranked second in edible mushroom production.There were more than 7 million tons production volume of Auricularia heimuer in 2020.In the context of such a large volume of production,there are still problems such as incompatibility of varieties,substrates and management,and low degree of standardization.Therefore,this study selected shen 8-7,a lamellated Auricularia heimuer with strong adaptability and widespread cultivation,as the test material,screened the most suitable conventional wood chip formula and studied its physiological characteristics,and obtained the following results:Changes in the proportion of wood chips in the formula(78% ~ 93%)changed the content and ratio of carbon and nitrogen elements,the content and ratio of lignocellulose components after sterilization of the culture material,and affected the activities of laccase,lignin peroxidase,hexokinase,6-phosphoglucose dehydrogenase and α-ketoglutarate dehydrogenase during the germination period,which affected the degradation capacity of lignin and hemicellulose and further affected the mycelial The average growth rate,full bag time and rehydration rate of ear pieces were further affected.For every 5% increase in the wood chip ratio,the carbon content decreased by about 8 mg/g,the nitrogen content decreased by about 0.79 mg/g,and the glucose content decreased by about 0.62 g.The wood chip ratio increased from 78% to 93%,and the carbon to nitrogen ratio in the sterilized culture material increased from 52.63 : 1 to 67.81 : 1,the lignin content increased from 17.15% to 18.47%,and the cellulose content increased from The laccase activity decreased from 136.12 U/g to 87.71 U/g,the lignin peroxidase activity increased from 1.81 U/g to 4.51 U/g,the hexokinase activity decreased from 16.55 U/g to 10.55U/g,and the 6-phosphate glucose dehydrogenase activity decreased from 31.02 U/g to 16.04U/g,α-ketoglutarate dehydrogenase activity decreased from 32.71 U/g to 21.56 U/g,lignin degradation decreased from 60.43% to 54.15%,hemicellulose degradation decreased from65.58% to 59.94%,average mycelial growth rate decreased from 0.78 cm/d to The average growth rate of mycelium decreased from 0.78 cm/d to 0.69 cm/d,the full bag time extended from 32.05 to 36.44 days,and the rehydration rate of ear piece increased from 10.08 to 11.35 times.The addition of soybean flour to the formulation significantly increased the content of nitrogen in the culture material,changed the form of nitrogen present,reduced the carbon to nitrogen ratio,and significantly improved the utilization of carbon and nitrogen,the overall level of extracellular enzyme activity,lignocellulose degradation,the overall level of respiratory enzyme activity,glucose utilization,and glycogen storage capacity,thereby improving a variety of agronomic traits of Auricularia heimuer.Compared to the formulation with 88% wood chips addition and 10% bran addition,replacing only 2% bran with soybean flour increased the nitrogen content by 1.626 mg/g or 21.4%,decreased the carbon to nitrogen ratio to 50.33 : 1,increased carbon consumption by 15.14 mg/g or 3.31% and nitrogen consumption by 0.613mg/g or 3.65%,and the extracellular enzyme activity of The overall level of extracellular enzyme activity increased significantly,total lignin degradation increased by 3.07%,total cellulose degradation increased by 4.29%,total hemicellulose degradation increased by 2.08%,overall level of respiratory enzyme activity increased significantly,glucose utilization capacity increased significantly,glycogen storage increased by 27.67%,total weight loss of culture material increased by 7.21%,average dry weight per bag entity increased by 9.61 g,average fresh weight per The average fresh weight per bag was increased by 94.86 g,the biological efficiency was increased by 15.52%,the rehydration rate of ear piece was enhanced by 9.25%,the mycelial growth rate was accelerated by 0.05 cm/d,and the full bag time was shortened by2.09 days.Through the study of the physiological characteristics and various aspects of agronomic traits performance of Shen8-7 Auricularia heimuer variety,a holistic and systematic explanation was given to illustrate the degradation and utilization pattern of cultivation substrate and transformation mechanism of Auricularia heimuer,and a conventional wood chip formulation suitable for the application of hanging bag cultivation technology of this variety was screened,with 88% wood chips,10% wheat bran,2% soybean flour,1% lime and 1% gypsum.
