| To investigate the association of Melanocortin Receptorm(MC4R)and Myostatin(MSTN)gene polymorphisms with growth traits and body size indexes in Qianbei Ma Goat,this study used 598 Qianbei Ma Goat,a high-quality goat breed unique to the Guizhou plateau mountains,as test subjects.Three non-linear mathematical models,Logistic,Von Bertalanffy and Gompertz,were used to fit growth curves for body weight,and multivariate statistics and throughput coefficients were used to analyse body weight and body size.By direct sequencing and sequence alignment,the SNPs loci and genotype typing in MC4 R and MSTN genes were explored,while bioinformatics analysis of SNPs loci was carried out using relevant bioinformatics online analysis software to explore the association between SNPs loci and growth traits in Qianbei Ma Goat;RT-q PCR was used to detect the spatio-temporal expression pattern of MC4 R and MSTN genes.The results obtained through the implementation of this study are as follows:(1)Three nonlinear models were used to fit growth curves to the body weights of male and female Qianbei Ma Goat,and the Von Bertalanffy model was the best fit to the body weights of male and female goat.(2)Four SNPs were found in the MC4 R gene: g.59345469 C>A,g.59345871 A>C,g.59346263 A>G and g.59346773 T>C.All four mutant loci had significant effects on body weight and chest circumference of Qianbei Ma Goat.H5H5(AAAAGGCC)was the most favorable for the growth of Qianbei Ma Goat..(3)Three SNPs were identified in the MSTN gene: g.130227914 A > G,g.130228015 A> G and g.130228164 T >A.All three mutant loci had significant effects on body weight and tube circumference of Qianbei Ma Goat.H2H5(AGGGAT)was a favorable haplotype for growth.(4)Dual luciferase identified the most active promoter region of MC4 R gene-1118 to-839 bp. |
