480 Trionyx Sinensis(Chinese softshell turtles),whose initial weights are within range of(31.75±7.20)g,were divided into 4 groups with 3 repeats in each group and then fed with a trial feed including 0 mg/kg,50 mg/kg,100 mg/kg and 150 mg/kg sanguinarine respectively for 75 days in a bid to investigate the impacts of sanguinarine on their growth,body compositions,serum indexes and intestinal wall flora.In this way,we explored the effect of the addition of sanguinarine on the growth performance,meat flavor and immunity of T.sinensis,and provided a scientific basis for the healthy use of T.sinensis and the rational use of sanguinarine in T.sinensis feed.The test results of four groups,i.e.,the 0 mg/kg group,the 50 mg/kg group,the 100mg/kg group and the 150 mg/kg group,clearly show that the survival rate of the 50 mg/kg group and the 100 mg/kg group are significantly higher than that of the 150 mg/kg group and the 0 mg/kg group(P<0.05).The total amount the umami amino acids in turtle’s muscles in the 50 mg/kg group increases 1.7 mg/kg in contrast to the 0 mg/kg group,yet with OTU noticeable difference(P>0.05);but the 50 mg/kg group still has remarkably higher performance than the 150 mg/kg group(P<0.05)in this regard.Furthermore,along with the addition of sanguinarine in the feed,the ratio of crude protein(CP)in the body composition of T.sinensis tends to increase first and then decrease.Specifically,the ratio of crude protein of the 50 mg/kg group is significantly higher than that of its control group and the 150 mg/kg group(P<0.05).At the same time,the glucose content in the serum of T.sinensis tends to increase first and then decrease as well within the normal range with the addition of sanguinarine.The glucose content in the 50 mg/kg group and the 100 mg/kg group are significantly lower than that of the 150 mg/kg group and the 0 mg/kg group(P<0.05).Moreover,the activity of alkaline phosphatase(AKP)in the 50 mg/kg group is significantly higher than that of the 0 mg/kg group(P<0.05)and the activity of glutathione peroxidase(GSH-PX)in the 50 mg/kg group is significantly higher than that of the rest three groups(P<0.05).The activity of aspartate aminotransferase(AST)in the 50 mg/kg group and the100 mg/kg group,however,are significantly lower than that of the 0 mg/kg group.In summary,the results demonstrate that a moderate addition of sanguinarine in the feed of T.sinensis can significantly increase survival rates,reduce feed coefficients,improve nutrient levels and flavors,protect livers,enhance the metabolism of mineral elements such as calcium and phosphorus,and improve the ability to resist oxidation and the utilization rate of sugar.In this experiment,high-throughput sequencing method was used to analyze the structure of intestinal wall of Chinese sturgeon with different blood base dosages in the feed.The results showed that the OTUs of the intestinal flora in the control group were lower than other groups,and the control group.The Chao1 diversity index and ACE index were significantly lower than those in the 100 mg/kg group and the 150 mg/kg group(P<0.05).The Chao1 index and the ACE index showed a trend of increasing first and then decreasing slightly with the increase of the amount of hematine.The four groups of Shannon index and There was no significant difference in the Simpson index,and the overall increase with the addition of hemorrhine showed an upward trend.The PD index of the control group was significantly lower than that of the 100 mg/kg addition group.The analysis of Shannon index trend,PD index trend and Chao1 index and ACE index was basically the same.The results of PCo A analysis showed that the non-weighted Uni Frac distance showed a large distribution distance between the 100 mg/kg and 150 mg/kg groups and the control group and the 50 mg/kg sample.The weighted Uni Frac distance analysis showed that the sample distribution distance between the groups was far,but 100 mg.There were significant spacing between the /kg and 150 mg/kg groups and the control group and the 50 mg/kg sample.In terms of bacterial structure,the results of this experiment show that the intestinal tract of Chinese sturgeon is mainly composed of Proteobacteria,Actinobacteria,Chlamydiae,Bacteroidetes,Fusobacteria.These five categories of bacteria make up.With the addition of sulphate in the feed,the abundance of Proteobacteria,Cyanophyta and Phloem cinereus increased first and then increased with the addition of radixine,and the abundance of Bacteroidetes was compared.The control group showed a significant degree of change(P<0.05).Moreover,when the amount of sanguinarine was 100 mg/kg,the proportion of Helicobacteraceae was significantly reduced(P<0.05),and it had a strong inhibitory effect on Helicobacteraceae.The test results show that the addition of sanguinarine in the feed can increase the abundance of the microorganism to a certain extent,and can inhibit the growth of some bacteria.As all indicators analyzed above,it also suggested that it would be more suitable to add 50 mg/kg~100 mg/kg sanguinarine in the basic feed of T.sinensis.
