This study is to identify the effects of oligosaccharide supplemented in infant formula on growth performance,serum biochemical indices and nutrient utilization in SD rats as the model and the effects of oligosaccharide supplemented in dietary on growth performance,serum biochemical indices,nutrient utilization in weaned piglets and to investigate the effects of oligosaccharide supplemented in food and feed on growth performance and nutrient utilization in the neonate and mechanism research,and finally,to provide scientific basis for the production of safe,high-quality food and feed.1.Effects of oligosaccharide supplemented in infant formula on growth performance,serum biochemical indices,nutrient utilization in SD ratsAll 32 healthy SD rats with 15 days old(a week before weaning)with similar to the weight and randomly were divided into 2 groups,with 4 replicates per group and 4 SD rats per replicate.Control,rats in group A fed infant formula and diet;rats in group B&C fed infant formula supplemented oligosaccharide(FOS1.85%,GOS0.28%),the feeding test was carried for 28 days.The results showed as follows:1)Supplemention of oligosaccharide had no significantly effects on growth performance and organs coefficient of SD rats(P>0.05).2)Supplemention of oligosaccharide significantly decreased the TP and BUN content in serum(P<0.05),significantly increase the HDL-C content in serum(P<0.05),and tended to increase the CK content in serum(P=0.087);3)Supplemention of oligosaccharide had no significantly effects on the apparent digestibility of CP,EE and Ca(P>0.05),but significantly increase sucrase activity in posterior segment of SD rats intestine mucosa and lactase activity in anterius and posterior segment of SD rats intestine mucosa(P<0.05),had no significantly effects on expression of Lactase and Mgam genes in anterius and posterior segment of intestine mucosa(P>0.05)。These results suggest that the infant formula supplement with oligosaccharide prebiotics can decrease the TG,BUN content and increase the HDL-C and some amino acid content in serum,promote the digestibility of nutrients and have no effect of growth performance in SD rats.2.Effects of oligosaccharide supplemented in dietary on growth performance,serum biochemical indices,nutrient utilization in weaned pigletsA total of 60 healthy weaned piglets with 21 days old(a week before weaning)with similar to the weight and randomly divided into A,B,C 3 groups,with 5 replicates per group and 4 piglets per replicate.Control,piglets in group A fed base diet;piglets in group B&C fed base dietary supplemention of 0.20% and 0.25% oligosaccharide(35%XOS&IMO),the feeding test was carried for 28 days.The results showed as follows:1)Compared with control group,supplemention of 0.20% oligosaccharide significantly increased the TG content in serum,the apparent digestibility of DM,CP,EE,GE,sucrase activity in jejunum and ileum mucosa(P<0.05),highly significantly increased the apparent digestibility of Ca and Pi,lactase activity in duodenum mucosa(P<0.01),significantly decreased the F/G and the diarrhea rate(P<0.05),highly significantly decreased the ALT content in serum(P<0.01),tended to increase lactase activity in jejunum(P=0.076)and ileum mucosa(P=0.062);supplemention of 0.25% oligosaccharide significantly increased the liver coefficient,the GLU content in serum and the apparent digestibility of Pi(P<0.05),highly significantly increased the apparent digestibility of Ca(P<0.01),significantly decreased the TP content in serum(P<0.05),highly significantly decreased the diarrhea rate(P<0.01).2)Compared with 0.25% oligosaccharide group,supplemention of 0.20% oligosaccharide had no significantly effects on growth performance of piglet(P>0.05),tended to increase the spleen coefficient(P=0.099)and the kidney coefficient(P=0.071),significantly increased TG and TP content in serum,the apparent digestibility of DM,EE,lactase activity in ileum mucosa(P<0.05),highly significantly increase the apparent digestibility of Ca and lactase activity in duodenum and jejunum mucosa(P<0.01),decreased the GLU content in serum(P<0.05),tended to increase the apparent digestibility of CF(P=0.051).3)Supplemention of 0.20% oligosaccharide significantly increased microbiota class and species of dominant in the contents of ileum(P<0.05),tended to increase in phylum(P=0.062),and tended to decrease the function of glycan biosynthesis and metabolism to microbiota in ileum(P=0.064);supplemention of 0.20% oligosaccharide tended to increase the relative abundance of Firmicutes(P=0.066),Actinobacteria(P=0.058)and Megasphaera(P=0.050),tended to decrease the relative abundance of Proteobacteria(P=0.073)and Campylobacter(P=0.054)and significantly decreased the relative abundance of Spirochaetes,Tenericutes and Clostridium in cecum(P<0.05),dietary supplemention of 0.20% oligosaccharide significantly increase the function of enzyme families metabolism,metabolism of other amino acids,metabolism of terpenoids and polyketides,nucleotide metabolism and xenobiotics biodegradation and metabolism to microbiota in ileum(P<0.05)and tended to increase the function of carbohydrate metabolism in ileum(P=0.085).These results suggest that the diet supplement with oligosaccharide can increase the nutrient utilization,decrease the diarrhea rate,improve intestine mucosa morphology integrity and disaccharidease activity and the best supplement is 0.20%;supplement with 0.20% oligosaccharide can increase good bacteria in cecum like Lactobacillus、Megasphaera and decrease maleficent bacteria in cecum like Treponema、Campylobacter,and go on to change the function of metabolism.In conclusion,the infant formula and feed supplement with oligosaccharide can improve animal health,speed up the protein metabolism and improve the digestibility of nutrients in animals.
