| Artificial insemination technology has been widely used with the development of pig industry,but the effect of in vitro preservation of pig semen has always restricted the popularization of artificial insemination technology and the rapid development of socialized sperm supply.At present,the commonly used semen preservation techniques are cryopreservation and normal temperature preservation,and these two preservation methods have their own advantages and disadvantages.In recent years,it has been found that the cryopreservation technology of pig semen at 4-5℃ can not only effectively avoid the stress damage of pig sperm in ultra-low temperature environment,but also inhibit the metabolic activity of sperm during preservation,so as to achieve the purpose of prolonging the preservation time,so this preservation technology has attracted more and more attention.However,during the cryopreservation of pig semen,Reactive oxygen species(ROS)will be enriched in large quantities and react with plasma membrane,proteins in sperm and even DNA,which will cause oxidative stress of pig sperm and affect the structure and quality of pig sperm.Therefore,we hope to reduce the oxidative stress of ROS on pig sperm by adding an appropriate amount of antioxidants to the diluent of pig sperm,improve the quality of pig semen during low temperature storage,and better serve pig production.In this study,quercetin and resveratrol with different concentrations and compatibility combinations were added to the diluent of pig semen.By detecting the viability and vitality,plasma membrane integrity,mitochondrial activity,total antioxidant capacity,superoxide dismutase activity,malondialdehyde content and sperm movement parameters of pig semen,the improvement effects of quercetin and resveratrol on the quality of pig semen stored at low temperature were evaluated,and then the appropriate amount of related antioxidants in the diluent of pig semen was screened out.The results are as follows:1.Under the condition of low temperature storage,different concentrations of quercetin(0,12.5,25,50,100 and 200 mol/L)or resveratrol(0,12.5,25,50,100 and 200mol/L)were added to the diluent of pig semen,and the viability,plasma membrane integrity,mitochondrial activity and activity of pig sperm were detected.The results showed that adding 50 mol/L quercetin or 50 mol/L resveratrol alone could improve sperm viability,motility,exercise parameters,plasma membrane integrity,mitochondrial activity,T-AOC and SOD activity,reduce MDA content,and effectively improve the preservation effect of pig semen.2.Under the condition of low temperature storage,adding quercetin and resveratrol to the diluent of pig semen can improve the storage quality of pig semen,and the effect is better than adding quercetin and resveratrol alone.The combination of quercetin with concentration of 50 mol/L and resveratrol with concentration of 25 mol/L had the best effect.On the 7th day,the sperm viability,plasma membrane integrity and mitochondrial activity were 46.77%,32.32%,33.81% and 35.76% respectively,which were higher than those of the single addition group.The effect of semen preservation in the combined treatment group with 50 mol/L quercetin and 50 mol/L resveratrol was lower than that in the single treatment group.To sum up,the results of this study show that adding 50 mol/L quercetin or 50 mol/L resveratrol to the diluent of pig semen alone can improve the viability,vitality and various sports parameters of sperm,at the same time,improve the integrity of sperm plasma membrane,mitochondrial activity,T-AOC and SOD activity in semen serum,reduce MDA content and improve the effect of cryopreservation of pig semen.The combined addition of quercetin and resveratrol has a certain synergistic effect,which further improves the preservation effect of pig semen,and the combined addition of 50mol/L quercetin and 25 mol/L resveratrol has the best effect. |
