| The purpose of this paper is to substitute soybean lecithin for yolk in TRIS diluent and to study the effect of soybean lecithin powder diluent on low temperature preservation of sheep semen.The optimal dosage of soybean lecithin in sheep semen low temperature preservation diluent was determined by measuring sperm viability at different time and acrosome integrity rate.The effects of TRIS patent diluent and soybean lecithin diluent on low temperature preservation of sheep semen were evaluated by low temperature preservation of sperm viability at different time and acrosome integrity rate.Comparison of pregnancy rate of artificial insemination with in vitro fertilization: powder diluent and TRIS in order to obtain a soybean lecithin phosphate powder diluent which can be used to produce low temperature and high efficient storage of sheep semen,patent diluent can be used for low temperature preservation of sheep semen at low temperature.There are three parts in this study,the details of which are as follows:Experiment 1 : Effects of different concentrations of soybean lecithin diluent on low temperature preservation of sheep semen.The aim of this study was to determine the optimal dosage of soybean lecithin in sheep semen low temperature preservation diluent by replacing yolk in TRIS diluent with different concentration of soybean lecithin.(0%,0.25%,0.5%,0.75%,1.0%,1.25%)six concentrations of soybean lecithin were used to replace egg yolk in TRIS diluent for low temperature preservation of sheep semen.Sperm viability was measured at 0,1,4,7,10,12,18 days.Acrosome integrity rate.The results showed that: 1)the sperm motility of 0% group was significantly lower than that of other groups from day 1(P < 0.05),0.5%,0.75%.The sperm motility of 0.5%,1.0% and 1.25%groups were significantly higher than 1.0% and 1.25%(P < 0.05)on the 10 th day,and 0.5%,1.0%and 1.25%,respectively(P < 0.05),and that of 0.5% group was significantly higher than that of1.0%,1.25% group.2)the rate of acrosome integrity decreased slowly in each group,and there was no significant difference at each time point(P > 0.05).Experiment 2:Effect of powder diluent on low temperature preservation of sheep semen.In this experiment,the TRIS diluent was dried to make the diluted powder,and the effects of cryogenic drying powder and TRIS diluent were compared.The sperm viability and acrosome integrity rate were measured at 0,1,3,6,9,12 days after low temperature preservation,and the sperm viability and acrosome integrity rate were measured at 0,1,3,6,9,12 days after freeze-drying and low temperature preservation with TRIS diluent.Then adding 0.5% soybean lecithin diluent to make powder,comparing the preservation effect of 0.5% soybean lecithin diluent and TRIS diluent powder,the sperm viability and acrosome integrity rate were measured at 0,1,4,7,10,12 days.The results showed that: 1)TRIS diluent powder and TRIS diluent When sheep spermatozoa were preserved for a long period of time at low temperature,the sperm viability was more than 50% on the 9th day and the acrosome integrity rate was higher than 65%.There was no significant difference between TRIS diluent and TRIS diluent on sperm viability and acrosome integrity.2)0.5% soybean lecithin phosphor powder and TRIS diluent powder were used to cryopreserved sheep semen.There was no significant difference in the rate of acrosome integrity at each time point(P > 0.05).The sperm motility was higher than 50% after 10 days of storage.Experiment 3:Fertilization of sheep semen stored in different diluent at low temperature.In this experiment,0.5% soybean lecithin phosphor powder diluent,TRIS diluent powder and TRIS diluent were used for in vitro fertilization of ovine oocytes afterlow temperature preservation of sheep semen for 9 days,and the cleavage rate was counted.The natural estrus of estrus was artificially inseminated on the 9th day after low temperature preservation with TRIS diluent powder and TRIS diluent.Sheep semen was inseminated at low temperature for 10 days with 0.5% soybean lecithin diluted powder and TRIS diluent powder.The pregnancy rate of estrus ewe was counted by B-ultrasound after 40 days.The results showed that: 1)0.5% soybean lecithin diluted powder,The cleavage rate of ovine oocytes fertilized in vitro was 48.27%,50.89%and 51.47% on the 9th day after low temperature preservation of sheep semen with TRIS diluent powder and TRIS diluent respectively.There was no significant difference in the cleavage rate ofovine semen after low temperature preservation of the three diluents for 9 days.2)artificial insemination was performed between the diluted solution powder and the TRIS diluent on the9 th day after low temperature preservation,and there was no significant difference in the cleavage rate of ovine semen after low temperature preservation.The conception rate was 66.9%and 67.8%,respectively.There was no difference between the two groups(P > 0.05).3)0.5%soybean lecithin diluted powder and TRIS diluent powder at low temperature.The conception rate was 65.49% in 0.5% soybean lecithin diluted powder group and 67.65% in TRIS powder diluent group on the 10 th day after preservation for artificial insemination.There was no significant difference between the two groups(P > 0.05).In conclusion,0.5% soybean lecithin instead of yolk in TRIS diluent has the best effect on low temperature preservation of sheep semen.0.5% soybean lecithin diluent powder,TRIS diluent powder and TRIS diluent low temperature preservation sheep sperm viability,acrosome integrity rate,in vitro fertilization cleavage rate,artificial insemination rate are basically the same,we can see that the sperm viability,acrosome integrity rate,in vitro fertilization rate and artificial insemination rate are almost the same.0.5% soybean lecithin phosphor powder diluent can achieve the effect of long-term clow temperature preservation of sheep semen and can be used in practical production. |
