| Oxidative stress has a great impact on the development of animal husbandry.Anthocyanins have strong antioxidant activities,but the raw material cost of anthocyanins is high at present.Therefore,it is particularly important to explore efficient,safe,green and economic natural antioxidants to improve animal health and promote the development of animal husbandry.The purpose of this study is to provide reference for the development and utilization of anthocyanins in Spirodela polyrhiza.The main research contents and results are as follows:1.Optimization of anthocyanin extraction from Spirodela polyrhiza 7498In this study,the ultrasonic-assisted citric acid-ethanol solvent extraction method was used to extract anthocyanin from Spirodela polyrhiza 7498,and the p H differential method was used to quantitatively analyze anthocyanin of Spirodela polyrhiza 7498.The effects of factors such as solid-liquid ratio,ethanol concentration,p H value,extraction time and extraction temperature on the anthocyanin extraction rate of Spirodela polyrhiza 7498 were investigated by single factor test and PBD test.Finally,total anthocyanin(TA)was used as response value to optimize by response surface method,and the optimum conditions for anthocyanin extraction of Spirodela polyrhiza7498 were further determined.The results showed that the solid-liquid ratio had a significant effect on the anthocyanin extraction rate of Spirodela polyrhiza 7498,and the ethanol concentration and p H value had a significant effect on the anthocyanin extraction rate of Spirodela polyrhiza 7498.The optimal extraction parameters were as follows: solid-liquid ratio 1g:39 m L,p H 4.57,ethanol concentration 58%,extraction time 10 min,extraction temperature 50℃.Under these conditions,the actual anthocyanin extraction rate of Spirodela polyrhiza 7498 was 389 mg /100 g.2.Enrichment and purification of anthocyanins of Spirodela polyrhiza 7498In this study,macroporous adsorption resin chromatography was used to enrich and purify the anthocyanin crude extract of Spirodela polyrhiza 7498 which obtained in the previous chapter.Firstly,according to the static adsorption rate and desorption rate,the best adsorption ability and desorption ability were selected from AB-8,DM130,and X-5 macroporous adsorption resins.Then,the screened macroporous adsorption resin was used to carry out static and dynamic adsorption tests on the anthocyanin crude extract of multiple Spirodela polyrhiza 7498,respectively,to explore the influence of various factors on the adsorption rate and desorption rate in the static and dynamic tests.Finally,the influence of various factors on the purification of anthocyanin crude extract of Spirodela polyrhiza 7498 was analyzed,and the purification process parameters were optimized.The results showed that AB-8 macroporous adsorption resin had the best purification efficiency for anthocyanin of Spirodela polyrhiza 7498.The optimum static adsorption time is 6h,the optimum temperature is 30℃,and the optimum p H value is4.The optimal loading volume of dynamic adsorption was 50 m L,the optimal loading velocity was 40 m L/h,the optimal ethanol concentration was 60%,and the optimal desorption velocity was 50 m L/h.The final anthocyanin purity of Spirodela polyrhiza7498 was 61.8% under this purification condition.3.Effects of Spirodela polyrhiza 7498 anthocyanidins on antioxidant capacity in miceTo investigate the antioxidant effect of anthocyanins of Spirodela polyrhiza 7498 on C57BL/6 type mice.24 male C57BL/6 were randomly divided into 4 groups with 6mice in each group.The feed treatments were: Group C-maintenance feed;Group Dmaintenance feed and 10% Spirodela polyrhiza 7498;Group H-maintenance feedanthocyanin of Spirodela polyrhiza 7498 gavage 100mg/kg;Group L-maintenance feed-anthocyanin of Spirodela polyrhiza 7498 gavage 200mg/kg.The mice were fed for 28 days.After the experiment,the total antioxidant capacity(T-AOC),malondialdehyde(MDA)content,glutathione peroxidase(GSH-Px)and superoxide dismutase(SOD)activities of the heart,liver and kidney of mice were measured.The results showed that the anthocyanin of Spirodela polyrhiza 7498 could significantly increase the T-AOC,reduce the content of MDA in the kidney,and enhance the activities of GSH-Px and SOD in the heart,liver and kidney of mice.To sum up,the main conclusion of this study is that the anthocyanin of Spirodela polyrhiza 7498 is valuable for extraction.The anthocyanin of Spirodela polyrhiza 7498 can significantly enhance the antioxidant capacity of mice,protect mice organs from oxidative damage,and have good antioxidant capacity. |
