Objective:With the development of screening as well as diagnosis techniques and the application of related vaccines,the mortality rate of cervical cancer has decreased in recent years.However,in 2012,the Global Health Organization survey show that cervical cancer is still the 4th most common tumors in women worldwide.The developing countries account for 85 percent.The invasion and metastasis of cancer cells to pelvic lymph nodes and other distal tissues and organs are the main causes of failure in treatment,poor prognosis and even death in patients with cervical cancer.ADAMs is a kind of Ⅰ-type transmembrane molecule,sharing "metalloproteinase domain" with matrix metalloproteinases(MMPs).It has been found that there are more than 40 ADAMs family members,and most of them have proteolytic enzyme activity by which ADAMs extensively involved in the regulation of cell adhesion,cell fusion,cell migration,membrane protein shedding and proteolysis,signal transduction and so on.Therefore,it is important to understand the expression and clinical significance of ADAM10 in cervical cancer for diagnosis,treatment and prevention and treatment of tumor.Methods:1.20 specimens of cervical squamous cell carcinoma(without preoperative radiotherapy and chemotherapy)as well as 20 ones with benign gynecological diseases and negative cervical cancer screening were collected.Real-time quantitative RT-PCR(qRT-PCR)was used to measure the expression level of ADAM10.Analysis the difference of expression between cervical cancer and non-cancer tissues2.Immunohistochemical method was used to detect the expression of ADAM10 protein in 67 cases of cervical squamous carcinoma,36 cases of cervical intraepithelial neoplasia(CIN)and 12 cases of normal cervical tissues.The relationship between the expression of ADAM10 protein and clinicopathological parameters was analyzed.Analysis of the relationship between the expression of ADAM10 protein and the survival of patients by Kaplan-Meier analysis3.The expression of squamous cell carcinoma antigen(SCC-Ag)in the serum of 84 patients with cervical squamous cell carcinoma was detected by immunoradiometric assay and its correlation with ADAM10 was analyzed.Results:The expression of ADAM10 mRNA in cervical cancer was significantly higher than that in non-cancerous tissues.From normal squamous epithelium of cervix to CIN to squamous cell carcinoma,the expression of ADAM10 was significantly increased.The expression of ADAM10 was not correlated with age,tissue differentiation and lesion size,but positively correlated with FIGO stage and lymph node metastasis and the difference was statistically significant(P<0.05).ADAM10 was positively correlated with the expression of SCC-Ag in patients with cervical squamous cell carcinoma with statistical significance(P<0.05)Conclusion:The expression level of ADAM10 is significantly increased with the development of cervical squamous cell carcinoma,and is closely related to FIGO stage and lymph node metastasis.Therefore,ADAM10 may be used as a biological indicator potential for the diagnosis of cervical squamous cell carcinoma and predicting and evaluating of therapeutic efficacy.
