| Bombyx batryticatus is a valuable medicine composed of dried silkworm larvae(Bombyx mori larvae)that were stiffened by infection with Beauveria bassiana.It has many applications in traditional Chinese medicine.This study is designed to address the inhibitory effect of different bioactive constituents isolated from Bombyx batryticatus ethanol extract on Gastric Cancer SGC-7901 cells.The B.batryticatus ethanol extract fractionation was resulted in four fractions,which are dichloromethane,ethyl acetate,n-butanol and aqueous fraction.The MTT test indicated that amongst the 4 factions,n-butanol fraction has a significant inhibition on SGC-7901,in addition to that,the inhibitory activity correlated with the treatment concentration and time.On the other hand,n-butanol fraction has no effect on the healthy normal cells BHK-21.The observed cells morphology when SCG-7901 treated with n-butanol fraction showed that,the majority of treated cells suffered from distortion,being detached,became smaller,round and grew slowly.When,the control cells grew normally.The apoptosis morphology under fluorescent confocal microscope detected that by increasing n-butanol concentration and incubation period cells were suffered from more distortion in both cytomembrane and nucleus.Cell apoptosis analysis determined that the apoptotic cells were increased 16.24% and 22.08% when n-butanol fraction was used at 60 and 200 μg/m L,respectively.Cell cycle analysis showed that,the cell number has been increased in G1 and G2 phases after the treatment with n-butanol.While the gene expression analysis explained that n-butanol fraction treatment could significantly up regulate the expression of P21 and Bax,and down regulate the Bcl2 expression at both RNA and protein levels.The results suggest that the n-butanol fraction might perform an important role in tumor growth inhibition by inducing SGC-7901 cells apoptosis through regulation of Bcl2/ Bax mediated apoptotic pathway.The liquid chromatography-Mass spectrometry analysis revealed that,the n-butanol fraction mainly composes of Amino acids,Alkanolamine,Phenols,Sugars,Falvonoids,Nucleosides and Organic acids.However,the key ingredients of anticancer activity in B.batryticatus were still not clear and more study was needed to identify them and their function. |
