Objective:To study the ?-solamarine bases induced cytotoxicity and apoptosis mechanisms of gastric cancer SGC-7901 cells.Method:Gastric cancer SGC-7901 cells were subcultured,the study was divided into negative control groups,different concentrations of ?-solamarine administered groups and positive control groups of HCPT.Using the MTT and SRB assay to detect ?-solamarine role in the proliferation of human gastric cancer SGC-7901 cells after 48h;Using the fluorescence microscopy to observe the effect of different concentrations of ?-solamarine bases on the cell cell apoptosis and cell cycle changes of different concentrations of p-solamarine;Fluo-3/AM mark,confocal laser scanning microscopy to detect changes in cell Ca2+ concentration;ELISA assay detect the activity of Caspase-3 and Caspase-12 in cell.Results:?-solamarine on SGC-7901 cell morphology after staining Hoechst33258;PI staining and flow cytometry detection SGC-7901 proliferation was inhibited in a dose-dependent manner;Under the fluorescence microscope found ?-solamarine effect on SGC-7901 cells appear obvious apoptosis after form;?-solamarine on SGC-7901 cell cycle arrest in G2/M phase;With the increase of drug delivery dosage,apoptosis rate are dose dependent growth;?-solamarine can raise concentration of Ca2+ in SGC-7901 cells;the activity of Caspase-3 and Caspase-12 in cells were increased.Conclusion:?-solamarine can inhibit the SGC-7901 cell proliferation,and block the eell cycle in G2/M phase,by influencing the calcium ion concentration,increase the activity of Caspase-3 and Caspase-12 in endoplasmic reticulum pathway lead to tumor cell apoptosis.
