| Objective:Acute lymphocytic leukemia(ALL)is a group of hematological malignancies originating from hematopoietic stem/progenitor cells,which are divided into T-cell ALL and B-cell ALL.lncRNA is a non-coding RNA with length greater than 200 nucleotides,which widely exists in Eukaryotes.It has been suggested that lncRNA plays an important role in various normal cellular activities and pathogenesis of diseases,such as the epigenetic regulation,cell cycle and cell differentiation.More and more studies focus on this filed.Here we try to detect the gene expression level of HOX-AS2,a lncRNA located in7p1.52,in bone marrow specimens of patients with acute lymphoblastic leukemia and the controls with non-malignant hematological diseases.Also,we analyze the correlation between the expression level and clinical features including the prognosis.Methods:Bone marrow species from 46 patients with acute lymphocytic leukemia and 32 cases with non-malignant hematological diseases as controls were collected.Gene expression levels of HOXA-AS2 were detected by RT-PCR.The median age of acute lymphoblastic leukemia group was 38 years(17-63 years)including 24 males and 22 females;the median age of the control group was 35 years(18-66 years)including 17 males and 15 females.The cutoff value for higher expression group and lower expression group was median gene expression level of HOX-AS2.Correlation of HOX-AS2 gene expression level with 37 cases of clinical characteristics were analyzed.Significant difference was defined as P<0.05 by Stata analysis software and online Bioinformatics analysis was performed on Star Base.Results:1.The mean expression of HOXA-AS2 was significantly higher in the patients with acute lymphoblastic leukemia(p = 0.0434).2.The complete remission(CR)rate was significantly lower in the patients with higher HOXA-AS2 expression(p = 0.0428).3.The relapse free survival(RFS)in patients with higher expression of Lnc RNA HOXA-AS2 was significantly lower(p = 0.0428).4.Bioinformatics analysis indicated that micro RNA-520c-3p was one of the potential target gene of lncRNA HOXA-AS2,which might be involved in the pathogenesis of ALL.Conclusion:The gene expression level of lncRNA HOXA-AS2 is higher in the patients with acute lymphoblastic leukemia,which is associated with lower CR rate and shorter RFS,which indicates that HOXA-AS2 could be used as biomarker predicting the poor prognosis of the disease. |
