ObjectiveAcute myeloid leukemia(AML)is a group of highly heterogeneous hematological tumor diseases,most common in adult leukemia.Although the cure rate has increased in recent years,drug resistance and relapse of the disease are the obstacles of AML treatment.Research reported that abnormal gene expression in AML affects disease progression and treatment and was related to prognosis.In the early stage of this study,using bioinformatics technology to analyze the ONCOMINE database,it was found that the GPX1,SOCS5 and IL7 were abnormally expressed in AML,and were related to the prognosis of AML.Combined with the literature,GPX1,SOCS5 and IL7 were abnormally expressed in a variety of tumors,and participate in the occurrence and development of tumors,and were related to the drug resistance and prognosis of tumor treatment.SOCS5 and IL7 played an important role in the JAK/STAT signaling pathway.But the expression in AML and its clinical significance are not clear.Therefore,this study analyzes the relationship between the expression level of GPX1,SOCS5 and IL7 in bone marrow cells of newly diagnosed patients with AML(non-M3)and analyzed the relationship between their expression levels and the clinical characteristics laboratory indicators and the efficacy of induced chemotherapy of AML patients,for the purpose of analysis the clinical significance of GPX1,SOCS5 and IL7 in AML.MethodsFirst,bone marrow samples were collected from 66 AML newly diagnosed patients and 32 healthy controls from the same period.And then mononuclear cells were extracted from the bone marrow samples by density gradient centrifugation.Subsequently real-time fluorescence quantitative PCR(RT-PCR)was used to detect the expression level of GPX1,SOCS5 and IL7 in the bone marrow cells of the study subjects.At the same time,these patients with AML were followed up to collect clinical data of AML patients.Eventually the relationships were analyzed between the clinical data and GPX1,SOCS5 and IL7 expression level of AML patients by using the SPSS statistical software.Results1.Used RT-PCR technology to detect the expression levels of GPX1,SOCS5 and IL7 in 66 newly diagnosed AML patients and 32 healthy persons in control group.The result showed that,compared with the control group,GPX1 expression in AML was significantly down-regulated,P<0.05.The expression of GPX1 in AML was 0.52 times that of the control group.While the expression levels of SOCS5 and IL7 were not significantly different between the two groups,P>0.05.2.Analyzed the relationship between GPX1 and clinical features of AML.The results showed that GPX1 expression was related to CEBPA-mutant gene and FAB typing,P=0.037 and P=0.006,respectively.3.Analyzed the relationship between GPX1 and the efficacy of induction chemotherapy in patients with AML,the result showed that the expression difference of GPX1 between CR group and refractory group was statistically significant,P=0.029.4.Analyzed the relationship between GPX1 expression level and the prognosis of AML patients,the results showed that GPX1 expression level was not related to the survival prognosis of AML patients.Cox regression analysis of the factors affecting the prognosis of AML showed that transplantation(P=0.044,HR=0.115,95%CI:0.014-0.947)and induction chemotherapy were independent good factors for the prognosis of AML patients(P<0.001,HR=0.178,95%CI:0.073-0.436).Conclusions1.The expression of GPX1 was significantly down-regulated in patients with AML.Compared with the healthy control group,the expressions of SOCS5 and IL7 in AML was not significant.2.GPX1 expression was correlated with CEBPA mutant gene,FAB typing and the effect of induction chemotherapy of AML patients.GPX1 expression is not correlated with prognosis and survival of AML patients.The efficacy of transplantation and induction chemotherapy was an independent good factor for the prognosis of AML.
