| Objective: To explore the protective effect of traditional Chinese Korean medicine Jiawei Rushen Decoction on ethanol-induced chronic gastric mucosal injury,and to provide experimental andtheoreticalbasis forrelatedresearch and clinicalapplication.Methods: SD male rats were randomly divided into six groups: normal group,model group,Sucralfate group,and Jiawei Rushen Decoction group(high-dose group,medium-dose group,low-dose group),then 8 in each group.The model was established by ethanol gavage,and the chronic gastric mucosal injury model was formed after 12 weeks.After successful modeling,the control group and model group were given normal saline(2ml/200g),Suc group(2.06ml/kg),JRD high-dose group(10.68g/kg),JRD medium-dose group(5.34g/kg),JRD low-dose group(2.67g/kg).After 4 weeks of continuous intragastric administration,fasting without water for 24 hours and taking serum and gastric tissue.The contents of epidermal growth factor(EGF),prostaglandin E2(PGE2),endothelial-1(ET-1)and interleukin-6(IL-6)in serum of rats were determined by enzyme-linked immunosorbent assay,the observation of pathological changes of gastric mucosa by hematoxylin-eosin(HE)staning method,and the expression of cyclooxygenase-2(COX-2)protein in gastrictissuewasobserved byimmunohistochemical staining.Results: 1.As a result of the determination of EGF content showed that compared with the model group,the content of each JRD group and Suc group were increased,and the difference was statistically significant(P<0.05).Compared with Suc group,the EGF content of the JRD high-dose group was significantly higher(P<0.05).2.The results of PGE2 determination showed that compared with the model group,the content of each JRD treatment group and the Suc group were increased,and the difference was statistically significant(P<0.05).Compared with the Suc group,the content of PGE2 in JRD high-dose group was significantly higher(P<0.05).3.The results of ET-1determination showed that compared with the model group,the content of each JRD group and Suc group were decreased,and the difference was statistically significant(P<0.05).Compared with Suc group,the content of ET-1 in JRD high-dose group was higher,but the difference was not statistically significant(P>0.05).4.The results of IL-6determination showed that compared with the model group,the content of each JRD group and Suc group were decreased,and the difference was statistically significant(P<0.05).Compared with Suc group,the content of IL-6 in JRD high-dose group was higher,but the difference was not statistically significant(P>0.05).5.Through HE staining,it was found that the gastric mucosal in each JRD group recovered well,only a small amount of mucosal congestion was seen,and no obvious inflammatory cell infiltration was found,so each JRD group had the effect of repairing the gastric mucosa.6.Immunohistochemical staining showed that compare with the model group,the expression of COX-2 in all medication administration groups were significantly reduced,especially at high dose of JRD,the difference was statistically significant(P<0.05).Conclusion: Traditional Chinese Korean medicine Jiawei Rushen Decoction caneffectively repairthechronic gastricmucosal injury inducedbyethanol inrats,itsmechanismmayberelated to increasing thecontentofepidermal growthfactor and prostaglandin E2,reducing thecontentof endothelium-1andinterleukin-6,and reducing the expression of COX-2 protein ingastric tissue. |
