Study On The Effect Of The Anti-Gastric Ulcer Effect Of Traditional Chinese Korean Medicine Jiaweiqiqi Decotion Combined With Omeprazole And Its Mechanism

Objective:To explore the mechanism of traditional Chinese Korean medicine JiaweiQiqi decoction combined with omeprazole in gastric ulcer,this study provide experimental and theoretical basis for clinical application.Methods:The clean-grade SD rats were randomly divided into five groups:normal control group(Control),model group(Model),omeprazole group(Ome),Jiawei Qiqi decoction group(JQD),and Jiawei Qiqi decoction combined with omeprazole group(JQD+Ome),8 rats in each group.The model was made by intragastric administration of anhydrous ethanol(1ml/200g).After successful intragastric administration,the Normal group and Control group were given 2 times of normal saline(1ml/100g),Ome group(2.1mg/kg)and JQD group(3g/kg)were given drugs and normal saline(1ml/100g),JQD+Ome group were given drugs and normal saline(lml/100g).The interval between the two gavages was one hour for 14 days.The contents of pepsin,NO,and bFGF in serum samples of rats were detected by ELISA.Additionally,the ulcer index was measured,and the inhibition rate of the ulcer was calculated.The pathological morphology and changes of gastric mucosa were observed by HE staining.The expression of bFGF protein in gastric tissues was observed by immunohistochemical(IHC)staining.Results:1.The results of pepsin determination showed that the pepsin content in the JQD+Ome group were lower than that of Model group(P<0.05),bearing statistical significance.2.The results of NO determination showed that the content of NO in the JQD+Ome group was significantly higher than that in the JQD group(P<0.05),the difference was statistically significant.3.The results of bFGF measurement showed that the bFGF in the JQD+Ome group was higher than that in the JQD group and Ome group(P<0.05),bearing statistical significance.4.The results of ulcer index measurement indicate that,the ulcer index of JQD+Ome group was lower than that of Ome group and JQD group(P<0.05),bearing statistical significance.5.HE staining showed that the structure of gastric mucosa in the JQD+Ome group was completely repaired,and the inflammatory exudate disappeared.6.Th e IHC staining showed that the expression of bFGF in the Model group was weak positive,in each treatment group,the expression of bFGF was higher than that in the Model group,and a large number of positive cells were also found.The expression of bFGF in the JQD+Ome group was higher than that in the JQD group and Ome group(P<0.05),bearing statistical significance.Conclusion:Traditional Chinese Korean medicine JiaweiQiqi decoction combined with omeprazole can decrease ulcer indexpromote,repairment of damaged gastric mucosa,and promote the healing of gastric ulcer rats.The mechanism may related to the decrease of serum pepsin,the increase of serum nitric oxide and bFGF,also to increase of bFGF protein expression.
