Objective:To observe the levels of resistin and inflammatory factors in the serum of patients with diabetic retinopathy(DR),including interleukin-1 beta(IL-1β),IL-6,IL-8,IL-12,tumor necrosis factor-alpha(TNF-α),C-reactive protein(CRP),vascular endothelial growth factor(VEGF),intercellular adhesion molecule-1(intercellular adhesion molecule-1,ICAM-1)and vascular cell adhesion molecule-1(VCAM-1),analyze the correlation between serum resistin and the above inflammatory factors,to further explore the effect of serum resistin on the pathogenesis of DR.Method:A total of 104 subjects were selected and divided into four groups: 35 patients with proliferative diabetic retinopathy(PDR),17 patients with nonproliferative diabetic retinopathy(NPDR),30 patients with diabetes mellitus(DM)without DR,and 22 healthy controls.The concentrations of resistin,IL-1β,IL-6,IL-8,IL-12,TNF-α,CRP,VEGF,ICAM-1 and VCAM-1 in peripheral blood of the subjects were examined by enzyme Linked immunosorbent assay(ELISA),to compare the levels of resistin and the above-mentioned inflammatory factors among the groups.The correlations between resistin and these inflammatory factors were analyzed,and the effect of resistin on DR development was analyzed by regression analysis.Result:The levels of serum resistin,IL-1β,IL-6,IL-8,IL-12,CRP,VEGF,ICAM-1,and VCAM-1 in the PDR patient group,NPDR patient group and DM patient group were all significantly higher than those in the healthy control group(all P<0.01).The levels of resistin,IL-1β,VEGF,ICAM-1 and VCAM-1 in PDR patients were significantly higher than those in DM patients(all P<0.01),however,there were no significant differences between the PDR group and the NPDR group,and between the NPDR group and the DM group(all P>0.05).The serum levels of IL-6,IL-8 in PDR group were significantly higher than those in NPDR group and DM group(P=0.030,P<0.001;P=0.005,P<0.001),but there was no significant difference between the NPDR group and the DM group(both P>0.999).Compared with healthy controls,serum TNF-α levels were significantly increased in the PDR patient group and NPDR patient group(both P<0.001),furthermore,serum TNF-α levels in PDR patients and NPDR patients were significantly higher than those in DM patients(P< 0.001;P =0.023),but,there were no significant differences between the DM group and the HC group or between the PDR group and the NPDR group(P=0.178;P = 0.495).The serum CRP levels in the PDR patient group were significantly higher than that in the NPDR patient group and the DM patient group(both P<0.001),at the same time,the NPDR patient group was significantly higher than those in the DM patient group(P<0.001).There were no significant differences in serum IL-12 levels among the PDR patient group,the NPDR patient group and the DM patient group(all P>0.999).The serum resistin levels of DR patients had no significant correlation with IL-1β,IL-6,IL-8,IL-12,TNF-α,CRP,VEGF,ICAM-1 and VCAM-1(all P>0.05).Logistic regression analysis showed that serum TNF-α,CRP,ICAM-1 levels were independently associated with the occurrence and progression of DR in DM patients,whereas the levels of resistin,IL-1β,IL-6,IL-8,VEGF and VCAM-1 were not independently correlated with the development of DR(all P>0.05).Conclusion:1.The levels of resistin,IL-1β,IL-6,IL-8,TNF-α,CRP,VEGF,ICAM-1 and VCAM-1 in patients with DR were significantly increased.But there are no significant correlations between resistin levels and the above inflammatory factors in DR patients.2.TNF-α,CRP and ICAM-1 may be independently associated with the development of DR;however,serum resistin is not an independent risk factor for DR development.
