Objective :Immune inflammatory response is one of the important pathogenesis of diabetic retinopathy(DR)and plays an important role in the development of DR.Cytokines and inflammatory factors play a key role in the regulation of inflammatory responses.Vascular endothelial growth factor(VEGF)is currently the most important angiogenic growth factor in patients with proliferative diabetic retinopathy(PDR),,but clinical anti-VEGF treatment can not prevent the development of PDR conditions,,the occurrence and development of PDR are influenced and regulated by a variety of inflammatory and growth factors.Interleukin-19(IL-19)as a cytokine closely related to the pathogenesis of inflammation and autoimmune diseases.This study examined the expression levels of IL-19 and VEGF in serum and vitreous fluid in patients with PDR,the role of IL-19 and VEGF in the pathogenesis of PDR was preliminarily explored to provide a new idea for the prevention and treatment of PDR.Methods: A total of 77 patients(77 eyes)who underwent vitrectomy were selected in ophthalmology department of our hospital.37 patients(37 eyes)with proliferative diabetic retinopathy(PDR)as the experimental group(PDR group),40 patients(40 eyes)with rhegmatogenous retinal detachment(RRD)as the control group.The gender and age of all subjects were recorded,and the duration of diabetes was recorded and the levels of fasting blood glucose(FBG)? glycosylated hemoglobin(HbAlc)were detected in PDR group.Enzyme-linked immunosorbent assy(ELISA)was used to detect the concentrations of IL-19 and VEGF in the serum and vitreous fluid of the two groups,the serum and vitreous IL-19 and VEGF levels in the two groups were compared.Results: 1.The serum levels of IL-19 and VEGF in the PDR group were significantly higher than those in the control group(IL-19:40.15±8.01 pg/ml vs 26.15±5.95 pg/ml;VEGF:72.50±17.61 pg/ml vs 20.52±8.04 pg/ml),the differences were statistically significant(t=8.747,16.437,P<0.01).2.The vitreous levels of IL-19 and VEGF in the PDR group were significantly higher than those in the control group(IL-19:53.18±13.41 pg/ml vs.25.61±9.26 pg/ml;VEGF:972.17±386.00 pg/ml vs 21.58±7.04 pg/ml),the differences were statistically significant(t=10.417,14.978,P<0.01).3.The levels of IL-19 and VEGF in the vitreous of PDR group were higher than those in serum(IL-19:53.18±13.41 pg/ml vs 40.15±8.01 pg/ml;VEGF:972.17±386.00 pg/ml vs 72.50±17.61 pg/ml),the differences were statistically significant(all P<0.05).4.There was a positive correlation between IL-19 and VEGF in the serum of PDR group(r=0.394,p<0.05).There was a significant positive correlation between IL-19 and VEGF in the vitreous of PDR group(r=0.574,p<0.01).There was a positive correlation between serum IL-19 and vitreous IL-19 in PDR group(r=0.370,p<0.05).There was no significant correlation between serum VEGF and vitreous VEGF in PDR group(P>0.05).5.The VEGF in the vitreous of PDR group was positively correlated with FBG and HbAlc(r=0.361,0.672,all P<0.05),There was no significant correlation with the duration of diabetes(P>0.05);Serum VEGF and IL-19 in PDR group had no significant correlation with diabetes course?FBG and HbAlc(all P> 0.05).Conclusion:1.IL-19 and VEGF are highly expressed in patients with PDR,IL-19 and VEGF may be involved in the pathogenesis of PDR.2.The expression of IL-19 is closely related to VEGF,IL-19 may be involved in the occurrence and development of neovascularization in PDR.
