Expression And Clinical Significance Of CEP55 And PLK1 In Ovarian Serous Carcinoma

Objective:By studying the expression of centrosomal protein 55 k Da(CEP55)and Polo-like kinase 1(PLK1)in ovarian serous carcinoma,the relationship between the expression of the two proteins and the clinicopathological characteristics of patients and the correlation between the two proteins were analyzed.It provides a certain experimental basis and basis for the prognosis evaluation and targeted therapy of ovarian serous carcinoma patients.Methods:A total of 50 ovarian serous carcinoma specimens diagnosed by pathology in the First Hospital of Shanxi Medical University from January 2020 to August 2021 were collected,including 20 cases of low-grade serous carcinoma and 30 cases of high-grade serous carcinoma.50 cases of normal fimbria tissues of fallopian tube were used as the control group.The expressions of CEP55 and PLK1 in ovarian serous carcinoma and normal fimbria tissues of fallopian tube were detected by immunohistochemical staining combined with Western blot.SPSS26.0 statistical software was used to analyze the correlation of CEP55 and PLK1 protein expressions and their relationship with the clinicopathological characteristics of the patients.Results:1.Immunohistochemical results showed that the high expression rates of CEP55 in ovarian serous carcinoma and normal fimbria tissues of fallopian tube were 60%(30/50)and 14%(7/50),respectively,with a statistically significant difference(P<0.001).The positive rate of PLK1 in ovarian serous carcinoma was 64%(32/50)compared with 10%(5/50)in normal fimbria tissues of fallopian tube(P<0.001).2.Western blot results indicated that the relative expression of CEP55 protein in ovarian serous carcinoma(1.043±0.162)was higher than that in normal fimbria tissues of fallopian tube(0.457±0.061),and the difference was statistically significant(P<0.05).The relative expression of PLK1 protein in ovarian serous carcinoma(0.648±0.073)was significantly different from that in normal fimbria tissues of fallopian tube(0.177±0.022)(P<0.05).3.The results of immunohistochemistry and Western blot showed that the expressions of CEP55 and PLK1 in high-grade serous carcinoma were higher than those in low-grade serous carcinoma and normal fimbria tissues of fallopian tube,with a statistically significant difference(P<0.001).However,there were no significant difference in the expressions of CEP55 and PLK1 between low-grade serous carcinoma and normal fimbria tissues of fallopian tube(P>0.05).4.The expression of CEP55 was significantly correlated with the FIGO stage,histological grade and lymph node metastasis of ovarian serous carcinoma(P<0.05),but was not significantly related to age(P>0.05).The expression of PLK1 was associated with the histological grade of ovarian serous carcinoma(P<0.05),but had no significant correlation with FIGO stage,age and lymph node metastasis(P>0.05).5.The expressions of CEP55 and PLK1 were positively correlated in ovarian serous carcinoma(r=0.408,P<0.05).Conclusion:The protein expressions of CEP55 and PLK1 in ovarian serous carcinoma were significantly increased and positively correlated.It can be speculated that CEP55 and PLK1 play a certain role in ovarian serous carcinoma.The high expression rate of CEP55 in high-grade serous carcinoma is higher than that in low-grade serous carcinoma.At the same time,for ovarian serous carcinoma patients with higher FIGO stage and lymph node metastasis,the high protein expression rate of CEP55 also increased,which suggests that CEP55 may promote tumor cell proliferation,invasion and metastasis through certain molecular signaling pathways.The positive rate of PLK1 in high-grade serous carcinoma was significantly higher than that in low-grade serous carcinoma,and the prognosis of low-grade serous carcinoma was significantly better than that in high-grade serous carcinoma.Therefore,the higher expression rate of PLK1 may indirectly predict the poor prognosis of patients.
