| Objective: To investigate the protein expression of centromere protein U(CENPU)and polo-like kinase 1(PLK1)in tissues of epithelial ovarian cancer(EOC)and their clinical significance in EOC.Research Methods: The key gene CENPU was screened by bioinformatics method,and the expression of CENPU and PLK1 protein in ovarian epithelial tumors and the clinical significance and correlation between the two in EOC tissues were analyzed by immunohistochemical staining.Results: The EOC datasets GSE27651,GSE40595,GSE54388 were downloaded from the GEO database,and 1517 co-expression differential genes were screened,including 923 down-regulated and 594 up-regulated genes,and GO and KEGG enrichment analysis was performed on the differentially expressed genes,and in biological processes the genes were mainly enriched such as organelle division,nuclear division,chromosome separation,nuclear chromosome separation,mitosis division,spindle,cell-cell junction and other cellular components,tubulin binding,Microtubule combination and other molecular functions are mainly enriched in cell cycle,proteoglycans,Hedgehog signaling pathways and other signaling pathways in the cell cycle,and 21 key genes were screened,of which 12 were related to the overall survival rate of EOC patients,CENPU was highly expressed in EOC tissues,and its high expression reduced the survival rate of EOC patients,and it was found that CENPU was closely related to PLK1,and immunohistochemical results showed that the expression of PLK1 and CENPU proteins in EOC tissues was elevated.Compared with the ovarian epithelial junction and benign group,the difference was statistically significant(P<0.05),the positive expression rate of CENPU protein in EOC tissue was related to the degree of tissue differentiation,clinical stage,and ascites,and the difference was statistically significant(P<0.05),and there was no significant correlation with lymph node metastasis,and the positive expression rate of PLK1 protein in EOC tissue was related to clinical stage and tissue differentiation.The difference was statistically significant(P<0.05)and was not significantly correlated with the presence or absence of ascites and lymph node metastasis.CENPU was positively correlated with the expression of PLK1 protein in EOC tissues(r=0.446,P<0.05).Conclusions: The high expression of CENPU and PLK1 protein in EOC tissues is positively correlated,which has an important impact on the survival prognosis of EOC patients,and its targeted therapy may provide new ideas for the treatment of EOC. |
