Objective:Based on network pharmacology,this thesis analyzes the mechanism of Shengyang Jieyu Decoction in the treatment of depression,and further observes the clinical efficacy of Shengyang Jieyu Decoction in the treatment of depression through randomized controlled trials,in order to provide reference for traditional Chinese medicine therapy of depression in the future.Method:The gene information corresponding to the chemical components,potential targets and protein targets of the traditional Chinese medicines in Shengyang Jieyu Decoction was obtained by searching the Sym Map database and Uniprot database one by one,and the network diagram of "traditional Chinese medicine-component-target" was constructed by Sytoscape 3.6.1.The disease targets related to depression were retrieved from the Sym Map database.To obtain the target of Shengyang Jieyu Decoction in the treatment of depression.From January 2021 to October 2021,66 patients with mild and moderate depression(depression/depression)of Yang deficiency type were randomly divided into two groups,33 in each group,who were treated by Gu Shisong,chief physician,in the outpatient department of psychosomatic department of the Affiliated Hospital of Tianjin Academy of Traditional Chinese Medicine.The control group was treated with sertraline tablets,The study group was treated with Shengyang Jieyu Decoction on the basis of the control group,the treatment cycle was 8 weeks,and the efficacy of Hamilton scale score,self-rating depression scale score and TCM syndrome integral scale score before and after treatment was compared between the two groups.Result:1.The study shows that Shengyang Jieyu Decoction has the characteristics of multi-component,multi-target and multi-way in the treatment of depression.2.There were 60 cases of clinical observation,30 cases in the control group and 30 cases in the study group.3.The HAMD scores of the two groups before and after treatment were compared within the group,and repeated test analysis of variance was used to show that the HAMD scores changed over time,with a significant downward trend,and the downward trend of the study group was more obvious than that of the control group.The total effective rate of the study group was 93.33%,and the total effective rate of the control group was 70.00%.The comparison of the HAMD score between the two groups was P < 0.05.The difference was statistically significant.4.The SDS scores of the two groups before and after treatment were compared within the group,and repeated test analysis of variance was used to show that the SDS scores changed over time,with a significant downward trend,and the downward trend in the study group was more obvious than that in the control group.The total effective rate of the study group was 93.33%,and the total effective rate of the control group was 63.33%.The SDS scores of the two groups were significantly different(P < 0.05).The difference was statistically significant.5.The scores of TCM syndrome integral scale before and after treatment in the two groups were compared within the group,and repeated test analysis of variance was used to show that the scores of TCM syndrome integral scale changed over time,with a significant downward trend,and the downward trend in the study group was more obvious than that in the control group.Two groups of patients before and after treatment of TCM syndrome integral scale score efficacy comparison,the total effective rate of the study group was96.67%,the total effective rate of the control group was 66.67%,the two groups of TCM syndrome integral scale score efficacy,P < 0.05,the difference was statistically significant.6.The study group had fewer side effects,milder symptoms and higher safety than the control group.Conclusion:1.Shengyang Jieyu Decoction in the treatment of depression involves multiple components,multiple targets and multiple pathways.Combined with network pharmacology analysis,Shengyang Jieyu Decoction in the treatment of depression may be related to reducing inflammatory response,regulating immune stress,regulating cell differentiation,regulating nervous and endocrine metabolism.Associated with TNF signaling pathway,human cytomegalovirus infection,AGE-RAGE signaling pathway in diabetic complications,Kaposi’s sarcoma-associated herpesvirus infection,prolactin signaling pathway.2.Through clinical research and observation,we can see that Shengyang Jieyu Decoction combined with sertraline tablets has better efficacy and higher safety in the treatment of depression of Yang deficiency and liver depression than single sertraline tablets.3.Director Gu Shisong believes that depression,loss of interest,laziness and tiredness,loss of appetite and other manifestations belong to "Yin",so he believes that depression is caused by Yang deficiency of the body,the main location of the disease is in the liver,heart and kidney,tired,treatment with warming and tonifying Yang Qi as the first priority,regulating Qi and opening depression should be carried out from beginning to end,treatment should be early,and prevention before disease is the main treatment principle.Establish the treatment method of tonifying Yang qi and soothing liver qi.Shengyang Jieyu Decoction has a significant effect in the treatment of mild and moderate depression,and can significantly improve the accompanying symptoms of depression,with less adverse reactions and safe clinical application.
