| Objective:To explore the mechanism of berberine in reducing intestinal mucosal barrier defect caused by colitis by inhibiting pyroapoptosis.Methods:1.Firstly,in vivo model of mouse colitis induced by sodium Dextran sulfate(DSS)was used to construct ulcerative colitis model in mice induced by sodium Dextran sulfate(DSS);2.After 3d adaptive feeding,C57BL/6J mice were randomly divided into 7 groups by random number table method :(1)Normal control group(Normal);(2)DSS group(DSS);(3)DSS+ berberine group(D+BBR);(4)DSS+ Mir-103 A-3p AGO Mir-(synthesis of overexpressed Mir-mimics)group(D+ agomiR-103a-3p);(5)DSS+ agomiR-103a-3p negative control(D + agomiR-103a-3P-NC),(6)DSS+BBR+Ant-agomiR-103a-3p(miRNA inhibitor)(D+B+Ant-agomiR-103a-3p),(7)DSS+BBR+miR-103a-3p AntagomiR-103a-3p negative control(D+B+Ant ago mir-103a-3P-NC).3.Body weight,fecal consistency,fecal occult blood and general physical symptoms of mice in each group were recorded every day.The permeability of intestinal epithelium was evaluated,the pathological structure of colon tissue was observed by HE staining,the degree of cell apoptosis in colon tissue was evaluated,the serum inflammatory factors were detected and the target protein bands were visualized and analyzed by western blot,so as to evaluate cell pyrosis.Results:Berberine alleviates cell scortosis and intestinal mucosal barrier defects stimulated by colitis by elevated Mir-103A-3p or inhibitory BRD4.Berberine can elevate Mir-103A-3p targeting BRD4.Inhibition of Mir-103A-3p or enhancement of BRD4 reversed the therapeutic effect of Berberine on colitis injury.Berberine inhibits activation of the Wnt/β-catenin pathway through the Mir-103A-3p /BRD4 axis.Berberine inhibits activation of the Wnt/β-catenin pathway by regulating the Mir-103A-3p /BRD4 axis,thereby reducing cell scortosis and intestinal mucosal barrier defects stimulated by colitis.Conclusion:By regulating Mir-103A-3p /BRD4 axis,berberine inhibits the activation of Wnt/β-catenin pathway and inhibits cell scortosis,thus reducing the intestinal mucosal barrier defect induced by colitis. |
