OBJECTIVE:To study the effect of raspberry safflower compatibility on serum glucose and lipid metabolism and sex hormone levels in PCOS-IR model rats,and to detect PI3K/Akt signaling pathway related factors to explore its mechanism of action,and to provide a scientific basis for its clinical treatment of PCOS-IR.METHODS:A total of 102 6-week-old SD female rats with a body weight(160±5g)of SPF grade were selected and divided into control group and model group according to the random number table method,including 10 rats in the control group and 92 rats in the model group.PCOS model rats were constructed by letrozole+high-fat diet,and the estrous cycle and HOMA-IR>2.8 of the rats were observed for two consecutive cycles as a sign of successful modeling.Finally,60 PCOS-IR rat models were successfully induced(success rate 65.22%).According to the same method as before,the rats with successful modeling were divided into the model group,the second group A,the drug pair group,the raspberry group,the safflower group,and the blocker group(PI3K inhibitor),with n=10 rats in each group.only.The intervention was carried out according to the treatment mode of each group for a total of 30 days.The morphological changes of rat ovary were observed by HE staining;FBG was measured by tail vein blood collection,serum TG,TC,T,E2,FSH and FINS of rats in each group were detected by ELISA,and then HOMA-IR was calculated;RT-PCR technology was used.The mRNA expression levels of PI3K and IRS-1 and the protein expressions of PI3K,Akt,IRS-1,FoxO3a and FoxO1 in ovarian tissue were detected by Western Blot technique.RESULTS:1.After modeling intervention,the rats in the control group had a complete estrous cycle,while the rats in the modeling group continued to be in estrus;the serum levels of FBG,FINS and HOMA-IR in the rats in the modeling group were significantly higher than those in the normal group(P<0.05).),and HOMA-IR>2.8;meanwhile,the body weight of the rats in the model group was significantly higher than that in the control group(P<0.05).After drug intervention,the body weight,ovarian mass,sex hormone level,and glucose and lipid metabolism levels of the rats in each treatment group were decreased to varying degrees compared with the model group(P<0.05),and the improvement in the metformin group and the drug pair group was particularly significant.The ovarian morphology of each treatment group was improved to varying degrees,especially in the two groups and the drug pair group,the ovarian morphology was significantly improved.2.The PI3K,IRS-1 mRNA levels and PI3K,Akt,IRS-1,FoxO1 protein expressions in ovarian tissue of the model group were significantly lower than those of the control group,and the FoxO3a protein expression level was higher than that of the control group.After intervention treatment,the expressions of PI3K,IRS-1 mRNA and IRS-1,PI3K,Akt,IRS-1,and FoxO1 protein in ovarian tissue of rats in the second group A and the drug pair group were significantly higher than those in the model group,and the expression level of FoxO3a protein was decreased.CONCLUSIONS:1.Kidney-tonifying herb raspberry combined with blood-activating anther,safflower,reduces IR in PCOS-IR rats by improving glucose and lipid metabolism.2.Kidney-tonifying herb raspberry combined with blood-activating anther safflower can improve reproductive function in PCOS-IR rats.3.Kidney-tonifying herb raspberry combined with blood-activating anther safflower can regulate reproductive endocrine hormone levels in PCOS-IR rats.4.Kidney-tonifying herb raspberry combined with blood-activating anther safflower can reduce IR and improve sex hormone secretion in PCOS-IR rats by activating PI3K/Akt signaling pathway.5.The combination of kidney-tonifying herb raspberry and blood-activating anther safflower can be further promoted in clinical treatment of PCOS-IR.
