Preliminary Study Of Pre-Treated With Benazepril Improving Collateral Circulation In Hypertensive Rats With Acute Cerebral Ischemia

Objective:(1)To assess the effect of pre-treated with angiotensin converting enzyme inhibitors(ACEI)on nerve function,infarct volume and cerebral collateral circulation in spontaneous hypertension rats(SHRs)with acute cerebral ischemia models and observe whether there were differences in different antihypertensive drugs((angiotensin receptor blockers,ARBs),calcium antagonists).Methods:32 male SHRs were randomly dividedd into 4 groups:group A was a blank control group(NS 2ml/d),group B was a benazepril group(benazepril 10mg/(kg.d)),group C is irbesartan group(irbesartan 50mg/(kg.d))and group D is amlodipine group(amlodipine 10mg/(kg.d)).All rats were fed drugs for 4 weeks,and then permanent middle cerebral artery occlusion(pMCAO)model was established.After modeling,the rats were treated with the preoperatively administered medication.Respectively,the cerebral blood flow and collateral circulation was observed by laser speckle imaging(LSI)at different time points(before modeling,15 minutes,24 hours,7 days after modeling),and 2 hours,24 hours,48 hours,72 hours,and 7 days after modeling,neurological function scores(Longa score)were assessed.Finally,the rats were sacrificed after LSI observation at 24h and 7d time point in every group and the whole brain tissues stained with 2,3,5-triphenyltetrazolium chloride(TTC).Results:(1)Observation of collateral on acute cerebral ischemic model in SHRs:in all groups,blood flow decreased in the occluded vascular area after modeling,and partial anastomotic vessels disappeared with time extension.The lateral branches were opened at each time point in each group,but the improvement was most obvious in group B and the pia meningeal branches were mainly opened at 15min,and neovascularization appeared 24h later.(2)Relative Cerebral Blood Flow(rCBF):rCBF in the observation area of rats decreased significantly after modeling.At each time node,rCBF value of group B,C and D showed an increasing trend compared with group A,but the difference was not statistically significant.(3)Longa score:after the successful modeling of rats,the scores of rats in each group were the highest at 2h and then decreased to varying degrees,but there was no statistically significant difference in the neurological function scores of each node among the groups.(4)Percentage of cerebral infarct volume with TTC staining:the percentage of 24h infarct volume in each group was larger than that on 7d.Compared with group A,the percentage of infarct volume in groups B,C and D was significantly decreased(P<0.05)at each time node,but there was no significant difference among drug intervention groups(P>0.05).Conclusion:Before modeling,treated with ACEI drugs,ARB and calcium antagonist can improve cerebral collateral circulation and reduce infarct volume in pMCAO model of SHRs,and ACEI drugs have a stronger potential to improve collateral circulation,but the effect on prognosis is inconclusive.
