ObjectiveBased on the clinical experience of mesalazine in the treatment of ulcerative colitis,to investigate the effect of mesalazine on NF-κB signaling pathway related factors in intestinal mucosal tissues of patients with ulcerative colitis,so as to provide basic supporting evidence for the effect and mechanism of drug treatment of ulcerative colitis.MethodsA total of 60 patients with ulcerative colitis admitted to Bishan Hospital of Traditional Chinese Medicine in Chongqing from January 2021 to January 2022 were selected as the research objects,and 60 subjects with normal microscopic examination during the same period were selected as the control group.The general sociological data and pathological characteristics of ulcerative colitis were analyzed in the two groups.The intestinal mucosal tissues of the two groups were collected,and immunohistochemistry,RT-PCR and Western-blot were used to determine the expression differences of key factors of NF-κB signaling pathway,i KKα,p-IKkα,IκB,p-IκB and p-NF-κB in the tissues.According to the clinical efficacy,the patients were divided into treatment effective group and treatment ineffective group.Intestinal mucosal tissues of the two groups were obtained.Immunohistochemistry,RT-PCR and Western-blot were used to determine the key factors of NF-κB signaling pathway in the tissues,including i KKα,p-i KKα,IκB,p-IκB.The expression of p-NF-κB was different.To determine the effect of mesalazine on NF-κB signaling pathway in mucosal tissues of patients with ulcerative colitis.Results(1)The differences in average age and sex ratio between the healthy control group and UC group were not statistically significant(P > 0.05);The VAS score and Mayo score of UC group were higher than those of Control group,and the ADL score was lower than that of UC group,with statistical significance(P<0.05).Compared with the Control group,the phosphorylation degree of NF-κB and the positive degree of p-NF-κB were increased,the phosphorylation of NF-κB and its binding factor IKKα was up-regulated,and the phosphorylation of NF-κB factor inhibitor protein IKKα was down-regulated in the colonic mucosal tissues of UC group(P<0.05).The expression of p-NF-κB in UC patients was positively correlated with the course of disease,VAS score and Mayo score,and negatively correlated with ADL score,and the difference was statistically significant(P<0.05).(2)After one course of treatment of UC,42 patients(70.00%)were effective and 18 patients(30.00%)were ineffective;The incidence of adverse reactions in effective group was lower than that in ineffective group,and the difference was statistically significant(P<0.05).Compared with the effective group,the phosphorylation degree of NF-κB and the positive degree of p-NF-κB were increased in the ineffective group,the phosphorylation of NF-κB and its binding factor IKKα was significantly up-regulated,while the phosphorylation of NF-κB factor inhibitor protein IKKα was down-regulated,and the differences were statistically significant(P<0.05).ConclusionsThe failure of mesalazine in the treatment of UC is closely related to the high expression of NF-κB in colon tissue.Mesalazine inhibits the development of UC by inhibiting the activity of NF-κB signaling pathway in colon intimal tissue,which provides some inspiration for clinical research.
