Objective: To evaluate the pharmacological activities of Ipomoea pescaprae aqueous extract(IPWE)against acute gouty arthritis(AGA)and rheumatoid arthritis(RA),and based on the theory of serum pharmacochemistry,using high performance liquid chromatography-mass spectrometry to clarify the pharmacodynamic material basis of Ipomoea pescaprae,which provided the basis for further study on the quality standard,in vivo process and mechanism of Ipomoea pes-caprae medicinal materials.Methods: 1 Study on the therapeutic effect of Ipomoea pes-caprae on AGA rat model: After 7 days of preventive administration,the rat model of AGA was induced by sodium urate and administered continuously for 2 days.The therapeutic effect of Ipomoea pes-caprae was evaluated by the observation of joint swelling,foot pad swelling,inflammatory scores,serum level of IL-1β,MIP-1α,MIP-2,RANTES,and synovial membrane histologic sections of AGA rats.2 Study on the therapeutic effect of Ipomoea pes-caprae on RA rat model:The therapeutic effect was evaluated by the observation of body weight,ankle swelling,plantar swelling,joint inflammation scores,organ index,pathological section of ankle joint,and serum level of PGE2,RF,CRP on RA rats induced by collagen-binding adjuvant.3 Study on the absorbed components of AGA rats treated with Ipomoea pes-caprae: 3.1 Qualitative analysis of the main chemical components of IPWE: LC-Q-TOF-MS/MS technology was used to qualitatively analyze the IPWE,which laid a foundation for the subsequent search for the absorbed components of Ipomoea pes-caprae.3.2 Study on the absorbed components of Ipomoea pes-caprae in normal rats: Based on the theory of serum pharmacochemistry to qualitatively analyzed the prototype components and metabolites of drug-containing plasma.3.3 Study on the absorbed components of Ipomoea pes-caprae in pathological rats: The plasma fingerprint of rats in normal blank group,pathological blank group and Ipomoea pes-caprae treatment group were compared,and the changes of absorbed components in AGA rats treated with Ipomoea pes-caprae were observed.Results: 1 Study on the therapeutic effect of Ipomoea pes-caprae on AGA rat model: Compared with the model group,the joint swelling index,foot pad thickness and joint inflammation index of IPWE group decreased significantly(P<0.01,or P<0.05)in AGA rats.The level of IL-1β,MIP-1α,and MIP-2expression in serum of the high and medium dose IPWE groups decreased significantly(P<0.05).There was a tendency to decrease the RANTES level in IPWE group,but there was no significant difference(P>0.05).Moreover,pathological sections of synovial tissue showed that the high and medium dose IPWE groups could reduce the infiltration of inflammatory cells.2 Study on the therapeutic effect of Ipomoea pes-caprae on RA rat model: Ipomoea pescaprae has a certain effect on relieving swelling and synovial inflammation in RA rats,and there were no significant difference in body weight,thymus and spleen index of each group after intervention that compared with the model group.There was a tendency to reduce the swelling degree of ankle joint and plantar joint in RA rats treated with IPWE,but there was no significant difference between treatment and model.Moreover,The high and middle dose IPWE groups could significantly reduce the joint inflammation index(P<0.05),the levels of RF in all IPWE groups(P<0.01),and the level of PGE2 and CRP in the high and middle dose IPWE groups(P<0.01,or P<0.05)were significantly lower than those in the model group.And pathological sections of the ankle joint showed that the pathological changes in the high and middle dose IPWE groups were relatively less than those in the model group.3 Study on the absorbed components of AGA rats treated with Ipomoea pes-caprae: 3.1Qualitative analysis of the main chemical components of IPWE: A total of 52 compounds were identified,including resin glycosides,flavonoids,organic acids,coumarins,nucleosides,amino acids,terpenes,lignans,phenolic acid glycosides and vitamins,among which organic acids and flavonoids were the main components.3.2 Study on the absorbed components of Ipomoea pescaprae in normal rats: Four components were identified as malic acid,caffeic acid,methyl caffeic acid and 4-hydroxybenzoic acid.3.3 Study on the absorbed components of Ipomoea pes-caprae in pathological rats: Four components were identified as caffeic acid,scopoletin,methyl caffeic acid and 4-hydroxybenzoic acid.Conclusion: 1 Ipomoea pes-caprae can significantly relieve joint swelling of AGA rats and inflammatory response,and inhibit the expression of IL-1β,MIP-1α,MIP-2.2 Ipomoea pes-caprae has significant therapeutic effect on RA rats,which is related to the reduction of PGE2,RF and CRP expression.3The main chemical components from Ipomoea pes-caprae were rapidly identified as organic acids and flavonoids by using LC-Q-TOF-MS/MS.4 LCQ-TOF-MS/MS was used to identify the blood components of Ipomoea pescaprae,which provided a reference for the basic research of its pharmacodynamic substances.
