| With the continuous improvement of people’s living standard,the people pay more and more attention to health problems.At present,the people still rely on western medicine for the treatment of difficult and complicated diseases,but the long-term use of western medicine will produce drug resistance and greater toxic side effects..In recent years,the people have gradually realized that traditional Chinese medicine has the advantages of remarkable curative effect and small toxic side effects,but it has not been able to push it to the international market.In order to promote the internationalization of traditional Chinese medicine and benefit the community with a shared future for mankind,the main scientific basis for its promotion and application will be to use modern scientific separation and analysis methods and clarify the material basis of traditional Chinese medicine’s efficacy.Astragalus membranaceus and Scutellaria baicalensis Georgi are recorded in the Chinese Pharmacopoeia.But the introduction of their medicinal components is relatively single,and Apium graveolens L has not yet been codified in the Chinese Pharmacopoeia.The Northern Shaanxi is located in the middle temperate zone,with a variable climate and large temperature differences between day and night.The growth of plants and their metabolites can vary depending on the climate.In order to fully explore the medicinal value of Astragalus membranaceus,Scutellaria baicalensis Georgi and Apium graveolens L planted in northern Shaanxi and their scientific and rational application,the chemical constituents and biological activities of the three medicinal plant resources were studied in this paper.Chapter 1 The chemical constituents and pharmacological actions of three medicinal plant resources were reviewed,which provided reference for the study of chemical constituents of Astragalus membranaceus,Scutellaria baicalensis Georgi and Apium graveolens L.Chapter 2 Extraction,separation and structural identification of chemical constituents from Astragalus membranaceus.The seven compounds were isolated and purified from the ethyl acetate extracted parts of 70%ethanol extract of Astragalus membranaceus using dextran gel LH-20 gel column chromatography and silica gel column chromatograph-y.The structures of these compounds were analyzed by using the physicochemical properties,Infrared Spectroscopy,Nuclear Magnetic Resonance and Mass Spectrometry data.Finally,the structure of the seven compounds was determined as:astragaloside IV,formononetin,calycosin,1-(4-hydroxyphenyl)-4-(2,4-hydroxyphenyl)-2-hydroxy-1,4-but anedione,(E)-4-methylcinnamic acid,quercetin and uracil.Chapter 3 Extraction,separation and structural identification of chemical constituents from Scutellaria baicalensis.The eight compounds were isolated and purified from the ethyl acetate part of the ethanolic extract of Scutellaria baicalensis using similar isolation means to that of Astragalus membranaceus.Finally,the structure of the seven compounds was determined as:baicalin,baicalein,wogonin,7-methoxychrysin,6-methoxyscutellarin,apigenin-7-methyl ether,lilacin and acanthin.Chapter 4 Extraction and isolation of chemical constituents and structure identification of Apium graveolens L.The ten compounds were isolated and purified from the ethyl acetate part an-d the aqueous part of the ethanolic extract of Apium graveolens L using similar is-olation means to that of Astragalus membranaceus.Finally,the structure of the se-ven compounds was determined as:palmitic acid,cinnamaldehyde,naringenin,Iso s-copoletin,xanthotoxin,6-methyl-5,7-dihydroxy Phthalide,2-phenylethyl-O-β-D-gl-ucopyranoside,apigenin-7-O-β-D-glucopyranoside,salidroside and D-apiose.Chapter 5 Cytotoxic activity test of monomeric compounds.Some monomeric compounds isolated from three medicinal plant resources were tested for their cytotoxic activity using MTT method.The test cells were:human lung cancer cells A549,human liver cancer cells BEL-7402 and human gastric cancer cellsSGC-7901.The results showed that compound Qi4 from Scutellaria baicalensis had strong cytotoxic activity against human lung cancer cells A549 with IC50value of 11.41μmol/L,and the IC50values for human liver cancer cells BEL-7402 and human gastric cancer cells SGC-7901 were 36.28μmol/L and 45.37μmol/L.The IC50values for compounds Hq3,Hq7 and Hq8 in Scutellaria baicalensis against human lung cancer cells A549 were 177.48μmol/m L,271.14μmol/m L,21.79μmol/m L.The compound Qc4 in Apium graveolens L had strong cytotoxic activity against human lung cancer cells A549,The IC50value is 6.68μmol/L.Chapter 6 Results and Prospects.A total of 25 compounds were isolated from Astragalus membranaceus,Scutellaria baicalensis Georg and Apium graveolens L.of northern Shanxi.Among them,one was a new compound and five had potential antitumor activity.These results lay a scientific foundation for the activity detection and action mechanism research in animals. |
