A Study On The Regularity Of Traditional Chinese Medicine In Treating Generalized Anxiety Disorder Based On Data Mining

Objective: Through collecting and sorting out the relevant literatures on the treatment of generalized anxiety disorder(GAD)with traditional Chinese medicine,with the help of traditional Chinese medicine inheritance auxiliary platform(V2.5),the medication regularity of GAD with traditional Chinese medicine was analyzed,and the commonly used combinations,potential new prescriptions and core prescriptions of GAD with traditional Chinese medicine were extracted.Based on network pharmacology,to explore the mechanism of action of traditional Chinese medicine in the treatment of GAD,and to provide new ideas for guiding clinical medication.Methods: CNKI,VIP database and Wanfang database were searched from April 2013 to April 2023.Published literatures related to traditional Chinese medicine with the first diagnosis of GAD were searched.Effective literatures were screened according to the inclusion and exclusion criteria.Relevant drugs were extracted and entered.Based on the traditional Chinese medicine inheritance auxiliary platform,the common combinations of traditional Chinese medicine,drug association rules,potential new prescriptions and core drugs were analyzed to summarize the medication rules.Based on the systematic pharmacology platform of traditional Chinese medicine,core drugs were searched,and oral availability and drug-like properties were set to screen active components and gene targets.The disease targets of GAD were searched in Gene Cards,Pharm GBK,and Dis Ge NET databases,and the core formula active component-disease target network was constructed in Cytoscape.Drug targets and disease targets were intersected and entered into the STRING platform to obtain a protein interaction(PPI)network,and then the Hub gene was selected using Cytoscape NCA for gene ontology pathway and Kyoto Gene and Genome Database pathway enrichment analysis.Results:1.A total of articles were included,and 199 complete prescriptions were extracted,involving 119 herbs.The four qi of the drug are mainly warm,cold and flat,the five flavors are mainly sweet,bitter and pungent,and the meridians are mainly heart,spleen and liver meridians,and the drugs with the highest frequency of use are Poria cocos,Glycyrrhiza uralensis,Bupleurum chinense,Zizyphus jujuba,and Radix Paeoniae Alba.2.In the association rule formula analysis,the most strongly related TCM combinations were "licorice – Poria cocos","Poria cocos – Polygala tenuifolia","Poria cocos – Zizyphus jujuba",and "licorice – Bupleurum chinense".Nine potential novel parties were obtained by prediction.Based on the degree of support,obtain the core formula composed of 11 herbs: Pinellia ternata,Citrus sinensis,Citrus aurantium,Bupleurum chinense,Paeonia lactiflora,Glycyrrhiza uralensis,Poria cocos,Zizyphus jujuba,Atractylodes macrocephala,Polygala tenuifolia,Angelica sinensis.3.There are 5698 GAD disease targets,573 core drug targets,and 458 active ingredients.Matching the disease target with the core prescription drug target,78 potential targets for disease treatment and 344 potential therapeutic compounds were obtained.After taking the intersection of drug targets and disease targets,309 points and 1354 edges were obtained in PPIs,and 22 Hub gene were selected using Cytoscape NC: MAPK1,HSP90AA1,PPARA,NCOA1,CTNNB1,VEGFA,IL6,NOS2,SP1,MAPK8,FOS,STAT3,RXRA,MAPK3,CREB1,EP300,EGFR,ESR1,JUN,MAPK14,CCND1,RELA.For the characterization parameters,the 12 most important active ingredients were screened: glacial acetic acid,quercetin,ursolic acid,kaempferol,eugenol,isoliquiritigenin,stigmasterol,levopethedrine,ephedrine,methylbutenol,oleic acid,isocorydaline.Conclusion:1.TCM treatment of GAD reflects the academic thoughts of combining cold and warm,pungent open and bitter down bearing and combined sweet,light and cold.2.The core drugs of traditional Chinese medicine for the treatment of GAD are: Pinellia ternata,Citrus sinensis,Zhishi,Bupleurum chinense,Paeonia lactiflora,Glycyrrhiza uralensis,Poria cocos,Zizyphus jujuba,Atractylodes macrocephala,Polygala tenuifolia,Angelica sinensis,Keel,and Oyster.3.Through network pharmacological studies,Glacial acetic acid,quercetin,ursolic acid,kaempferol,eugenol,isoliquiritigenin,etc.,the main active components of the core formula,may be through IL-6,ESR1,c-JUN,VEGF and other targets as well as Toll-like receptor signaling pathway,Fox O signaling pathway,TNF signaling pathway,response to reactive oxygen species,response to chemical stress,transcriptional regulatory complexes,vesicles,DNA-binding transcriptional activator activity,DNA-binding transcription factor binding,etc.,for anti-inflammatory,nerve regulation,anxiolysis,antidepressant effects,so as to achieve the purpose of treating GAD.
