Delphinium brunonianum Royle is a perennial herb of the genus Delphinium.It is mainly distributed in the southwest plateau of China,Nepal,Afghanistan and northern Pakistan.It grows on grassland or gravel between 4500 and 6000 meters above sea level.The main chemical components include alkaloids,anthraquinone glycosides,ascorbic acid,carbohydrates,coumarins,flavonoids,phenols,proteins and saponins.It is traditionally used to treat jaundice,fungal infection,influenza and snake bite.As a folk medicine,it often has the effects of clearing heat,detoxification,anti-inflammation and analgesia.In order to better develop and utilize the medicinal resources of Tibetan medicinal materials,in this paper,the separation,purification and structural identification of the chemical constituents of the flower were carried out.At the same time,the activity of the obtained compounds was screened by acetylcholinesterase,in order to find and clarify its inhibitory effect on acetylcholinesterase activity,and provide theoretical guidance for further study of this plant and the genus of Lepidium.(1)The types and proportion of compounds in the flowers were analyzed.Supercritical CO2 method was used to extract 5g of dry aerial part powder of L.japonica,and 0.0402g of extract was obtained.GC-MS was used to analyze the components of the extract obtained by supercritical CO2 method.The types and quantities of compounds contained in the flower were counted,and the proportion of various compounds was analyzed,so as to guide the follow-up work.(2)The chemical constituents of the flower were extracted.95%ethanol was used to extract the whole grass of Parrotbill at room temperature,and the total extract was obtained after vacuum drying and evaporation concentration.After the total extract was dispersed with water,it was extracted with petroleum ether,chloroform,ethyl acetate,n-butanol and other organic solvents to obtain different extraction sites.(3)The monomeric compound was isolated.A total of 11 monomer compounds were isolated and purified by thin layer chromatography,normal phase silica gel column chromatography and preparative chromatography.The structural formulas of 11 compounds were identified by NMR analysis of all the monomer compounds.Five compounds were identified asβ-sitosterol(1),Mannitol(2),Rutin(3),Naringenin(4)and T-ferulic acid(5).Six compounds were identified as Delsoline(6),Indole-3-carboxaldehyde(7),Lappaconitine(8),Eldeline(9),Benzoic acid(10)and P-hydroxybenzoic acid(11).Based on the analysis of previous research results,it was found that mannitol,delphinidin and lappaconitine were isolated from the flower of Delphinium brunonianum Royle for the first time.The research results enriched the study of compound components of Delphinium brunonianum Royle.(4)The activity of the monomeric compound was detected.The inhibitory effect of the obtained monomer compounds on acetylcholinesterase activity was determined by acetylcholinesterase activity inhibition experiment.The experimental results show that the three monomer compounds with better inhibitory effect on acetylcholinesterase activity are:rutin,naringenin and Eldeline.These three monomer compounds are expected to be developed into new cholinesterase inhibitors.
