ObjectiveTo extract the flavonoids portion of Prinsepia Utilis Royle, to separate the flavonoids monomer, and to study the bacteriostatic activity of the flavonoids portion, as to provide theoretical basis for further exploitation and utilization of the medicinal plants.MethodsTo get flavonoids portion by organic solvent extraction from Prinsepia Utilis Royle, and to measure the content of flavonoids of each extract layer material.Using column chromatography to separate and purify the part of the flavonoids portion of Prinsepia Utilis Royl to obtained compound monomer, and using mass spectrometry (MS), nuclear magnetic resonance spectrum (such as 1H-NMR and 13C-NMR) and other methods to identify the structure of compound monomer. Dilution method is used for the extract of ethyl acetate layer of Prinsepia Utilis Royle to study and research the bacteriostatic activity by the determination of minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of escherichia coli, staphylococcus aureus, pseudomonas aeruginosa, proteus, salmonella typhi, shigella dysenteriae, beta hemolytic streptococcus and candida albicans.ResultsThis experiment measured the content of flavonoids of each extract layer material, and the measured content of flavonoids of each extract layer material in total extract and petroleum ether, ethyl acetate, n-butanol and water were 3.00%,0.09%, 2.76%,0.09% and 0.03%.15 compounds were separated from Prinsepia Utilis Royle, and identified 10 compounds of them which were identified as eriodictyol (Ⅱ), kaempferol (Ⅳ), quercetin (Ⅴ), protocatechuic acid (Ⅵ), P-Sitosterol (Ⅶ), (+)-catechin (Ⅷ), margaric acid (Ⅺ), oleic acid (Ⅻ), ethyl-p-hydroxybenzoate (ⅩⅢ) and vanillic acid (ⅩⅣ). The MIC of staphylococcus aureus, pseudomonas aeruginosa, salmonella typhi, shigella dysenteriae, beta hemolytic streptococcus and candida albicans of the extract of ethyl acetate layer of Prinsepia Utilis Royle were 0.0110 g/ml, and there were not detected the MIC of escherichia coli and proteus. The MBC of escherichia coli, staphylococcus aureus, pseudomonas aeruginosa, proteus, salmonella typhi, shigella dysenteriae, beta hemolytic streptococcus and candida albicans of the extract of ethyl acetate layer of Prinsepia Utilis Royle were 0.1000 g/mL,0.0300 g/mL,0.0110 g/mL,0.0300 g/mL,0.0110 g/mL,0.0300 g/mL,0.0300 g/mL and 0.0300 g/mL.ConclusionThis experiment measured the content of flavonoids in each layer extraction of Prinsepia Utilis Royle in the first time, Separated and identified the 10 compounds to consist of 4 compounds of flavonoids,5 compounds of organic acids and a sterol compound, and the experimental result indicated that the extract of ethyl acetate layer of Prinsepia Utilis Royle has better antibacterial activity.
