| Objective:The aim of this study was to investigate the association of adenosine triphosphate binding cassette transporter G1(ABCG1)and transcription factor 7 analogue2(TCF7L2)gene single nucleotide polymorphisms and DNA methylation with type 2diabetes mellitus(T2DM)in Xinjiang Uygur population.Methods:A total of 320 T2 DM patients were selected as the case group and 332non-T2 DM patients were selected as the control group.Blood samples and clinical data of the subjects were collected.Minor Groove Binder(MGB)probe technology was used to detect gene polymorphism,and pyrosequencing was used to detect gene methylation.To analyze the association between TCF7L2 and ABCG1 genotypes and the risk of type 2diabetes.Rank sum test and chi-square test were used for univariate description,and Logistic regression was used to analyze the strength of association between T2 DM and genes.Results:1.(1)There were no significant differences in gender,age and sleep time between case group and control group(P > 0.05).However,there were differences in education level,household registration type,drinking,smoking and exercise between the two groups(P < 0.05).(2)There were significant differences in SBP,FPG,BMI,TG,TC,LDL-C and ALT levels between case group and control group(P < 0.05).There was no significant difference in waist circumference,DBP and AST(P > 0.05).(3)There were significant differences in hypertension,obesity,abdominal obesity,dyslipidemia and family history of T2 DM between the case group and the control group(P <0.05).(4)Logistic regression results showed that hypertension,smoking,family history of T2 DM,alcohol consumption,obesity,abdominal obesity and dyslipidemia were risk factors for T2DM(P < 0.05),while exercise and education level of high school or above were protective factors for T2DM(P < 0.05).2.There was a significant difference in the genotype of rs7901695 locus of TCF7L2 between the case group and the control group(P < 0.05).After adjusting covariates,the genotype of rs7901695 in the dominant model,additive model and recessive model of TCF7L2 gene was associated with the genetic susceptibility to T2DM(P < 0.05).3.After the intervention,SBP,DBP,TG,TC,LDL-C and other indicators were reduced in the case group and the control group.4.(1)There were significant differences in SBP,FPG,BMI,TG,TC,LDL-C and ALT levels between case group and control group(P < 0.05).There was no significant difference in waist circumference,DBP and AST(P > 0.05).(2)4.Correlation analysis between ABCG1 gene methylation and environmental factors showed that ABCG1 gene methylation was not associated with waist circumference,BMI,etc.,but was statistically associated with hypertension,obesity,dyslipidemia and fasting blood glucose.5.(1)There was no interaction between ABCG1 gene methylation and TCF7L2 gene polymorphism.(2)There were interactions between TCF7L2 rs7901695 and ABCG1cg06500161 with hypertension,dyslipidemia,abdominal obesity and obesity,and there were also interactions with alcohol consumption,smoking and exercise.Conclusion: TCF7L2 gene rs7901695 polymorphism is associated with T2 DM in Uygur.The methylation of ABCG1 gene cg06500161 may affect the development of T2 DM.The interaction of ABCG1 gene cg06500161 and TCF7L2 gene rs7901695 with environmental factors may increase the risk of T2 DM in Uygur population. |
