Objective: To explore whether the expressions of serum exosome derived miRNA-155-5p and miRNA-210-5p in severe preeclampsia,preeclampsia and normal pregnant women are related to the severity of the disease.Materials and methods: From 2021.01.01 to 2022.10.31,50 pregnant women with severe preeclampsia and preeclampsia(study group)who met the inclusion criteria were selected;50 pregnant women with normal delivery(control group)were selected.Serum exosomes were extracted by ultracentrifugation,total RNA from exosomes was extracted using miRNeasy Mini kit,c DNA was obtained by reverse transcription,and the expression levels of serum exosome-derived miRNA-155-5p and miRNA-210-5p were determined by RT-qPCR.SPSS26.0 statistical software was applied to analyze the data,and the Shapiro-Wilk test,one-way ANOVA,LSD method,quartiles and Kruskal-Wallis H test were used for statistical analysis,and Spearman’s correlation coefficient was used to test the correlation between variables.Results: 1.The differences were statistically significant when total serum RNA was compared between the three groups(P<0.05).2.A two-by-two comparison of miRNA-155-5p and miRNA-210-5p in the three groups: miRNA-155-5p and miRNA-210-5p levels in the preeclampsia group were 2.36 times and 5.26 times higher than those in the normal control group,respectively;The levels of miRNA-155-5p and miRNA-210-5p in the severe preeclampsia group were 5.96 and24.75 times higher than those in the normal control group,respectively;The levels of miRNA-155-5p and miRNA-210-5p in the severe preeclampsia group were 2.53 times and 4.70 times higher than those in the preeclampsia group,respectively.The differences were all statistically significant(P<0.05).3.There was a statistically significant positive correlation between miRNA-155-5p and miRNA-210-5p content and systolic blood pressure,mean arterial pressure and 24-hour urine protein in the severe preeclampsia group(P<0.05).Conclusions: Serum exosomal miRNA-155-5p and miRNA-210-5p are both highly expressed in preeclampsia,and both expressions increase with disease severity.It has some clinical applications.
