Objective: The purpose of this study is to explore the exosomal mi RNA from umbilical cord blood(UCB)in preeclampsia(PE)and normal pregnant women and the mechanism of key differential factor(mi R-125a-5p)working in preeclampsia,then determining the relationship between mi R-125a-5p and the pathogenesis of preeclampsia.Methods: 1.Human umbilical cord blood was collected from normotensive pregnancies and preeclampsia pregnancies who underwent caesarean delivery,and umbilical vein plasma exosomal mi RNA was extracted,then building a library and performing high throughput sequencing to obtain key factors(such as mi R-125a-5p),using quantitative real-time polymerase chain reaction(q RT-PCR)for verification.2.q RT-PCR and Western Blot was used to detect the expression of mi R-125a-5p and VEGFA(Vascular endothelial growth factor A)in placental tissues of normal and preeclampsia pregnant women.3.In vivo experiment: Human trophoblast cell line HTR8/SVneo was assigned into the negative control and mi R-125a-5p mimics.q RT-PCR and Western Blot were performed to identify the expressions of mi R-125a-5p and VEGFA.CCK8,flow cytometry,wound-healing and tube formation assays were used to analyze the effect of mi R-125a-5p on HTR8/SVneo proliferation,cycle distribution,cell migration and angiogenesis.Results:1.Compared with normal pregnant women,preeclampsia pregnant women showed higher expression of mi R-125a-5p in umbilical vein plasma exosomes;2.Compared with normal pregnant women,preeclampsia pregnant women showed higher expression of mi R-125a-5p in placental tissues and lower expression of VEGFA;higher expression of mi R-125a-5p in peripheral blood exosomes in late pregnancy;3.In HTR8/SVneo cells,compared with the negative control,the m RNA expression of VEGFA in the mi R-125a-5p mimics was lower,and the cell migration,proliferation and angiogenesis abilities were decreased;besides,more cells were arrested in the S stage.Conclusions: 1.The exosomal mi RNA expression profile between normal and preeclampsia pregnant women was significantly different;2.mi R-125a-5p can affect the migration and proliferation abilities of HTR8/Svneo cell,block the cell cycle and inhibit angiogenesis by regulating the expression of VEGFA,indicating that mi R-125a-5p could involve in the pathogenesis of preeclampsia and provide new thoughts for studying the mechanisms of preeclampsia.
