Preparation Of Zilian Cream And Preliminary Study On The Mechanism Of Wound Healing

Zilian ointment(formerly known as Huangjin Wanhong ointment)is a "star" product prepared by Yunnan Traditional Chinese Medicine Hospital.Composed of Zicao,Dihuang,Huanglian,Angelica sinensis,Polygonum cuspidatum,Scutellaria baicalensis,borneol,etc.,it has the effects of clearing heat and detoxifying,removing blood stasis and promoting regeneration,and reducing swelling and pain.Clinically,it is commonly used for burns,scalds,red buttocks in infants,skin ulcers,and infections.It has been applied in clinical practice for decades and has been highly praised by patients due to its wide and accurate therapeutic effects,convenient use,no obvious toxic side effects,and low price.Zilian ointment is an oily matrix ointment,which is relatively greasy,difficult to clean,easy to contaminate clothing,and has poor drug release and penetration effect.Objective This study considers making it an oil-in-water cream with appropriate viscosity,easy to apply to the skin and mucous membranes,uniform and delicate,lubricated without irritation,easy to clean,and does not stain clothing.Methods With the contents of emodin,berberine and baicalin as evaluation indicators,the extraction process of Zilian cream was optimized by orthogonal test,the optimal process conditions were determined,and the extraction method of Zilian cream was determined as follows: take prescription decoction pieces(except for shicao and borneol),add 70%ethanol to reflux and extract for 1.5 hours,decompress and recover ethanol and concentrate it to 1 g of the original drug liquid per ml.Take 6 times the amount of rapeseed oil from Zicao,heat until the oil boils,then add Zicao,extract for 5 minutes,filter with gauze,and obtain Zicao oil for later use.Using purple grass oil as the oil phase,distilled water as the water phase,self emulsifying matrix Tesose 63 as the emulsifier,and Labrifil M 1944 CS as the cosurfactant,the appearance uniformity,coating ductility,centrifugal stability,cold resistance,and heat resistance of the cream were evaluated as indicators.Single factor experiments and orthogonal experiments were used to investigate the optimal forming process of the formula.The emulsification time was 15 minutes,the emulsification temperature was 75 ℃,and the emulsification method was a mixture of water and oil,The dosage of Tefosse 63 is 10 g,and the dosage of Labrifil M 1944 CS is 5 g.A preliminary study was conducted on the quality of Zilian Cream,using L-shikonin and Shikonin as control materials,borneol as control materials,and Angelica sinensis as control materials.Thin layer chromatography was used to identify Shikonin,borneol,and Angelica sinensis in the prescription.The content of berberine in the preparation was determined by HPLC and methodological studies were conducted.Finally,the active ingredients of Zilian cream were obtained from the Systems pharmacology database and analysis platform of traditional Chinesemedicine(TCMSP)http://www.swisstargetprediction.ch/ Obtain the functional targets of each component and use the online Gene Cards database to screen potential targets for the active ingredients of Zilian Cream in treating wound healing.By combining the String database and Cytoscape’s Network Analyzer tool,identify potential target protein interactions.Using DAVID database,the potential target genes were analyzed by Gene Ontology(G0)classification enrichment analysis and genome(KEGG)pathway enrichment analysis.Result The results showed that 136 active ingredients were selected from Zilian Cream and acts on 81 targets,mainly through the regulation of 1.Erb B signaling pathway,2.PI3K-Akt signaling pathway,3.JAK-STAT signaling pathway and other signaling pathways to play a role in the treatment of skin wound healing.Conclusion Through the study of extraction process and molding process,the preparation method of Zilian Cream was determined as follows: take 15 g of Zicao oil,15 g of medicinal solution(1 g/m L),10 g of Tefose 63,and 5 g of Labrifil M 1944 CS,add distilled water to 100 g,heat in a 75 ℃ water bath for 15 minutes,stir while heating,cool to 50 ℃,add 1g of borneol powder,stir evenly,let cool,and solidify to obtain Zilian Cream.This preparation method uses purple grass oil as the oil phase,the extracted medicinal liquid as the water phase,and uses a self-emulsifying matrix for preparation.The process is simple and the stability is good.The identification method of Zicao,Danggui,and Borneol using thin-layer chromatography is simple,fast,negative control without interference,and specific;The content of berberine hydrochloride,the main active ingredient in Huanglian,the monarch,was determined by high performance liquid chromatography,Through the establishment of the detection method and methodological investigation,it was confirmed that this experimental method is simple,accurate,and has good repeatability.It can effectively control the quality of Zilian Cream,thus creating a prerequisite for the initial draft of the quality standard for Zilian Cream and for the development and application of new drugs in the formulation.A network pharmacology approach was applied to preliminarily predict the possible active ingredients,targets of action and signaling pathways of Zilian cream for wound healing.
