Objective: In this study,we selected Wenfei Xiaozhong Granules,a clinically effective empirical prescription based on the theory of "blood stasis and cold drink" from the research group ***,and used cigarette smoke combined with tracheal instillation of lipopolysaccharide to construct a chronic obstructive pulmonary disease.A rat model in the stable stage of obstructive pulmonary disease,to observe whether the prescription can inhibit the pathological changes of lung tissue in COPD rats,improve the systemic inflammatory factor levels of COPD rats,and observe the correlation of NF-κB pathway after the intervention of Wenfei Xiaozhong Granules.The expression changes at the protein level preliminarily explained the mechanism of action of Wenfei Xiaozhong Granules from the inflammatory mechanism of COPD.Methods:1.The COPD rat model was established by cigarette smoking combined with tracheal instillation of lipopolysaccharide.2.Monitor the general state performance and body weight of rats in different groups every day,and observe the changes of rats.3.Observing the lung inflammation and injury of different groups of rats by HE staining.4.Detect the contents of TNF-α and IL-6 inflammatory factors in arterial serum of rats in different groups by ELASA method5.Detect p-NF-κBp65 and NF-κBp65 in NF-κB pathway of rats in different groups by Western-blot method and p-IκBα,IκBα protein expressionResults:During the modeling process,the COPD rats may have obvious wheezing sounds,sluggish movement,yellow hair,listlessness,and slow weight gain.After four weeks of treatment with Wenfei Xiaozhong Granules,the symptoms of the rats were relieved.Compared with the model group,the body weight of rats in each administration group tended to increase.HE staining of pathological sections showed that the inflammation of the lung tissue of the rats was significantly improved after treatment.Compared with the model group,there were significant differences in IL-6 and TNF-α in arterial serum of the rats in the positive group,the middle-dose Wenfei Xiaoxiao group,and the high-dose Wenfei Xiaoxiao group(p<0.01).Compared with the model group,the expression levels of p-NF-κB,NF-κB,and p-IκB-α in the lung tissue of rats in each treatment group were all changed.High doses of Wenfei Xiaozhong Granules can significantly down-regulate the expressions of p-NF-κ B,NF-κ B,and p-I κ B-α in lung tissue of COPD rats.The expression of IκB protein was up-regulated(p<0.01),and the middle dose group of Wenfei Xiaozhong could down-regulate the expression of p-NF-κB protein in lung tissue of COPD rats.Up-regulate the expression of IκB protein(p<0.05).Conclusion: The above results show that Wenfei Xiaozhong Granules can inhibit the level of lung and systemic inflammation in chronic obstructive rats by inhibiting the NF-κB pathway.This study provides a theoretical basis and research basis for Wenfei Xiaozhong Granules to inhibit systemic inflammation in chronic obstructive pulmonary disease by regulating inflammation-related pathways...
