| Rationales: Aging is an inevitable and irresistible physiological change in the development of the body,which often accompanied by various degenerative diseases,such as heart disease,cancer,Parkinsonās disease and Alzheimerās disease,etc.The increasing prevalence of age-related diseases seriously affects the health of the elderly individuals.Therefore,delaying aging and improving the health of elderly people has been a major focus of the medical community.Persicae Semen has a wide range of pharmacological effects,such as antioxidation,enhancing immunity,anti-fibrosis,anti-inflammatory and neuroprotection,etc.It is often applied as soup in Chinese traditional medicine.Caenorhabditis elegans(C.elegans)is a species characterised by its small size,ease of culturing,and rapid reproduction.It has recently gained attention in anti-aging research,particularly after being isolated from long-lived strains.Thus,this study explores the anti-aging effect and mechanism of the water extract of Persicae Semen(WEPS)based on the C.elegans as the model organism.Objective: Evaluation of the efficacy of WEPS in delaying senescence of C.elegans by lifespan experiments and behavioral experiments;Searching for key target pathways for the anti-aging effects of WEPS through network pharmacology techniques;Validation of the anti-aging mechanism of WEPS by molecular biology techniques.Methods: This research utilizes C.elegans as a model organism to study the effects of WEPS on the longevity of C.elegans in natural aging,heat stress and paraquat damage models.In addition,we conduct the effects of WEPS on the fertility and motility of nematodes in naturing aging model.Furthermore,this study analyzes the mechanism of WEPS in delaying aging through network pharmacology.Finally,this study sheds light on the anti-aging mechanism of WEPS by testing the activities of SOD and CAT and the m RNA expressions of their related genes in C.elegans.Results: The results shows that longevity of C.elegans is prolonged when nematodes exposed to 0.5 mg/m L,2.5 mg/m L and 12.5 mg/m L of WEPS during the whole development stages.The administration group at 2.5 mg/m L proves to be the most effective in prolonging the lifespan of C.elegans in the natural aging,heat stress and paraquat damage models.Additionally,2.5 mg/m L of WEPS can improve the fecundity and motility of C.elegans in the natural aging model.Network pharmacology analysis reveals that Persicae Semen may affect the lifespan of organism by interfering with the antioxidant-related effects of the Lipid and atherosclerosis pathway and AGE-RAGE signaling pathway.Subsequent experiments examine the SOD and CAT genes expression levels and related enzyme activities in C.elegans.The results indicate that WEPS can increase the m RNA expressions of SOD and CAT genes,increase CAT and SOD activities in nematodes,and thereby reduce ROS content in C.elegans,delay nematode senescence.Conclusion: WEPS may enhance antioxidant activity and m RNA expression of related genes in C.elegans,thus enhancing the antioxidant effect and moderating oxidative damage in C.elegans,and ultimately achieving the ability to slow down C.elegans aging. |
