| Part1:Galectin-10 levels in serum and blister fluid of patients with bullous pemphigoid(BP)and their correlation with serum IgE and peripheral blood eosinophils.Objective: Comparison of Galectin-10 levels in blood and blister fluid and its correlation with serum IgE and peripheral blood eosinophils in patients with bullous pemphigoid.Methods: The Galectin-10 content in the serum of 14 bullous pemphigoid patients and 14 healthy subjects were detected by ELISA method,and the negative pressure formation of blister in 14 bullous pemphigoid patients and 14 vitiligo patients.The content of Galectin-10 in the blister fluid of the blisters was detected.The level of eosinophils in 14 patients with bullous pemphigoid was analyzed by sysmex XS-800 i automatic blood analyzer,and the serum total IgE level of patients with bullous pemphigoid was analyzed by cobas 8000 automatic chemiluminescent immunoassay analyzer,GraphPad Prism 8.0 was used to analyze the data with t-test and Pearson correlation coefficient.Results: Galectin-10 was positive in 100%(14/14)of BP patient sera and 100%(14/14)in blister fluid.The content of Galectin-10 in the serum of patients was 24.00±0.3739 ng/mL,and that of healthy subjects was 15.92±0.5526 ng/mL.The independent sample t test was performed between the two,and the difference was statistically significant(t=12.11,P<0.0001);the Galectin-10 content in the blister fluid of patients was 25.59±0.2930 ng/mL,and the Galectin-10 content in the blister fluid formed by negative pressure in patients with vitiligo was 18.22 ± 0.2691 ng/mL.The difference was statistically significant(t=18.52,P<0.0001).The Galectin-10 content in serum and blister fluid was analyzed by paired t test,and it was found that the content of blister fluid was slightly higher than that in serum,and the difference was statistically significant(t=8.341,P<0.0001).The correlation coefficient between the content of Galectin-10 in the patient’s serum and the count of eosinophils was0.7696,and the P value was 0.0013,so the content of Galectin-10 in the serum of the patient was moderately correlated with the count of eosinophils;The correlation coefficient between the Galectin-10 content and the total IgE is 0.6496,and the P value is 0.0119,so the Galectin-10 content in the patient’s serum is moderately correlated with the total IgE;The count correlation coefficient is 0.7890,and the P value is 0.0008,so the Galectin-10 content in the patient’s blister fluid is moderately correlated with the eosinophil count;the correlation coefficient between the Galectin-10 content in the patient’s blister fluid and the total IgE is0.6464,the P value was 0.0125,so the Galectin-10 content in the patient’s blister fluid was moderately correlated with the eosinophil count.Conclusions: This study found that the content of Galectin-10 is positive in both BP patients and normal controls,and the Galectin-10 content in the serum and blister fluid of patients is higher than that in the control group,and the Galectin-10 content in the serum and blister fluid of patients is different Significantly,the content of Galectin-10 in blister fluid was higher than that in serum;the peripheral level of Galectin-10 in patient serum and blister fluid was significantly correlated with eosinophil count;IgE was significantly correlated.Part2:Correlation between Galectin-10 levels in serum and blister fluid of patients with bullous pemphigoid and disease severity.Objective: To compare the correlation between serum and blister fluid levels of Galectin-10 in patients with bullous pemphigoid and their disease severity.Methods: The serum Galectin-10 content of 14 bullous pemphigoid patients was detected by ELISA method;the serum anti-BP180 antibody and anti-BP230 antibody of 14 bullous pemphigoid patients were detected by ELISA method,and the patients were evaluated.The severity of the disease was analyzed by Pearson correlation coefficient using GraphPad Prism v8.0.Results: The correlation coefficient between the content of Galectin-10 in serum of BP patients and anti-BP180 antibody was 0.5811,and the P value was 0.0293,so there was a moderate correlation between the content of Galectin-10 in serum and anti-BP180 antibody;The antibody correlation coefficient was 0.02358,and the P value was 0.9326,so the Galectin-10 content in the patient’s serum was not correlated with the BP230 antibody;The Galectin-10 content in blister fluid was moderately correlated with anti-BP180 antibody;the correlation coefficient between Galectin-10 content in blister fluid and anti-BP230 antibody was-0.03962,and the P value was 0.8946,so the Galectin-10 content in blister fluid was correlated with The anti-BP230 antibody was not correlated with the two;the correlation coefficient between the content of Galectin-10 in the serum of patients and the severity of the patient’s condition was 0.6722,and the P value was 0.0084,so the content of Galectin-10 in the serum was moderately correlated with the severity of the patient’s condition;The correlation coefficient between the Galectin-10 content in the blister fluid and the severity of the patient’s condition was 0.6411,and the P value was 0.0135,so the Galectin-10 content in the blister fluid was moderately correlated with the severity of the patient’s condition;The correlation coefficient of severity was 0.6211,and the P value was 0.0178,so there was a moderate correlation between the level of anti-BP180 antibody and the severity of the patient’s condition.Conclusions: This study found that the level of Galectin-10 in the patient’s serum and blister fluid was positively correlated with BP180 antibody;the level of Galectin-10 in the patient’s serum and blister fluid was not related to the anti-BP230 antibody;Severity is positively correlated.The patient’s anti-BP180 antibody level was also positively correlated with the severity of the
