Objective:Gastric cancer(GC)as the third largest cause of cancer-related death worldwide,has become one of the more serious disease burdens in China.Although the incidence of GC has declined by changing lifestyle and improving public health conditions,there is basically no treatment that can substantially improve the survival time of GC.The reason is that most of the patients have developed to the middle and late stages at the time of diagnosis.It is more important to explore markers for predicting tumor progression and patient survival.Based on the differential expression of the Special AT-rich Binding protein-1(SATB1)and the trefoil factor 3(TFF3)in GC,this study used a public database to explore the expression of SATB1 and TFF3 in gastric cancer and their correlation with the degree of gastric malignancy,and preliminarily verified pathologically,which provided an important experimental basis for the diagnosis and prognosis of gastric cancer.Methods:1.Pub Med,Webof Science,CNKI,Wanfang and VIPR databases were used to search the literatures published before December 30,2022,and screened according to the inclusion criteria.Odds ratio(OR)was used as a parameter to evaluate the expression level of SATB1 in GC tissues.Use STATA17.0software to integrate and complete the statistical analysis.In the analysis,when P< 0.05 or 95% CI did not include a value of 1,the combined results were considered to be statistically significant.2.TIMER2.0,GEPIA2 and UALCAN databases were used to explore the expression of TFF3 m RNA in GC,and GEPIA2 and Kaplan-Meier Plotter databases were used to analyze the prognostic value of TFF3 in GC.3.The expression of SATB1 and TFF3 in 40 patients with GC was detected by immunohistochemical method,and the relationship between the expression of SATB1 and TFF3 and clinicopathology and the correlation between them were analyzed.The chi-square test or Fisher’s exact method in SPSS23.0 was used for analysis,the correlation between SATB1 and TFF3 was analyzed by Spearman method,and P<0.05 was defined as meaningful.Results:1.Meta-analysis was used to analyze 14 studies(1052 patients).The results showed that the high expression of SATB1 was found in cancer tissue(OR=13.55,95%CI: 6.58-27.93,P<0.00001),old age(OR=1.89,95%CI: 1.45-2.47,P<0.0001),larger cancer focus(OR=1.37,95%CI: 1.00-1.88,P=0.049),lymphatic invasion(OR=3.69,95%CI: 2.39-5.71,P<0.0001),deeper invasion(OR=2.76,95%CI: 2.07-3.68,P<0.00001)and advanced stage(OR=3.78,95%CI: 2.79-5.11,P<0.0001),but not related to sex,location of cancer focus and degree of differentiation.2.The expression level of TFF3 m RNA in GC tissues was significantly higher than that in normal tissues,and the high expression of TFF3 m RNA in GC patients was related to cancer stage.In addition,survival analysis showed that the overall survival time of GC patients was associated with TFF3 m RNA expression.3.In 40 cases of GC,the positive rate of SATB1 was 62.5%(25/40).The expression of SATB1 was high in large cancer foci,lymph node metastasis,deep invasion and advanced stage(P<0.05).However,the sex,age,location of cancer focus,differentiation,vascular and nerve invasion were not related to the expression of SATB1(P>0.05).The positive expression rate of SATB1 was 70%(28/40)in 40 GC patients.The high expression of TFF3 was significantly correlated with deeper invasion and advanced stage of GC(P<0.05).No correlation was found between TFF3 expression and sex,age,size and location of cancer focus,differentiation,Lymphatic invasion,vascular and nerve invasion(P>0.05).Conclusions:1.SATB1 is highly expressed in GC,which is positively correlated with the development of tumor,so SATB1 can be used as a diagnostic marker for GC.2.Compared with normal tissues,the expression of TFF3 m RNA in GC tissues is increased,and it is significantly correlated with the overall survival time of GC.It can be used as a prognostic marker of GC.3.There is a positive correlation between the expression of SATB1 in GC and the expression of TFF3,so the diagnosis of combined SATB1 and TFF3 has a potential application prospect.Figure 17 Table 10 Reference 79...
